摘    要:目的:探讨成肌细胞冷冻复苏后,体外培养条件下的生物学特征。方法:将原代、2、4、6代成肌细胞液氮冻存,4周后复苏并进行体外培养,并与未冻存的相应各代次传代成肌细胞进行体外增殖特征比较;同时对传代和复苏成肌细胞的第2代细胞融合率进行了观察。结果:复苏后成肌细胞倍增时间、平顶期较对照组长;但成肌细胞形态、特性未见明显变化;复苏后成肌细胞融合率与传代细胞一致。结论:成肌细胞冻存后可以作为组织工程化面肌修复所需的种子细胞来源用于实验研究。

Abstract:Objective: The article is to investigate the biological characteristics of continuously subcultured frozen-thawed myoblasts of SD rats, and the possibility to be cryopreserved as seeding cells for facial muscle tissue engineering. Methods: Cryopreserved myoblasts of SD rats and the same sample without frozen as control group were subcultured in vitro. The growth of cells, rate of myotube formation (RMF) was used to evaluate the proliferative and defferentiation ability of myoblasts. Results: Within the 6 passages, the frozen-thawed myoblasts showed low proliferation. But it showed the same biological and morphological characteristics of myoblasts as control group. It also showed the same RMF between frozen-thawed myoblasts and control group. Conclusions: Cryopreserved myoblasts could be served as seeding cells in facial muscle tissue engineering.
