端粒酶hTRT mRNA在口腔癌前病变中的表达

摘    要:目的探讨端粒酶hTRT mRNA在口腔癌前病变中的表达相关性。方法采用原位杂交方法检测病变组织中hTRT mRNA的表达水平。其中非典型增生18例、单纯性增生5例、肿瘤周围邻近组织5例、正常口腔黏膜2例,共30例口腔病理标本。结果55.6%非典型增生(10/18)、20%单纯性增生(1/5)、20%肿瘤周围邻近组织(1/5)、0%正常口腔黏膜(0/2)端粒酶hTRT mRNA显示阳性表达。其中重度非典型增生与轻度及中度非典型增生之间端粒酶hTRT mRNA表达差异有显著性意义(P〈0.05)。结论端粒酶hTRT mRNA在口腔癌前病变中有表达。端粒酶活性的激活可能发生在口腔癌前病变晚期。其活性的激活是口腔癌前病变恶变及恶性肿瘤形成的重要因素。

Abstract:Objective To investigate the implications of hTRT mRNA expressing level in the tissues of oral precancerous lesion.Methods Eighteen cases with dysplasia in oral mucosa were included in this study with other 5 samples of simple hyperplasia in oral mucosa,5 samples of paracancerous oral tissue and 2 samples of healthy oral mucosa as controls.Results The positive expressing rates of hTRT mRNA were 55.6%(10/18),20.0%(1/5),20.0%(1/5)and 0(0/2)for the groups of dysplasia,simple hyperplasia,paracancerous tissue and healthy oral mucosa respectively,with very significant differences between the group of dysplasia and other groups of tissue samples.Furthermore,there was a significant difference in the expressing level of hTRT mRNA among the cases with severe degree of dy splasia and those with moderate degree of dysplasia(P<0.05).Conclusions The expressing level of hTRT mRNA has been significantly elevated in oral precancerous lesion and the activation of telomerase may occur at a late stage of this king of lesion.Then,it will promote developing of oral precancerous lesion into malignant change,even leading to a malignancy forming.
