Abstract:Objective To establish a thin layer chromatography(TLC)method and the content determination of phillyrin in Reduning Oral Liquid.Methods TLC method was used for qualitative identification of Flos Lonicerae,Folium Isatidis,Radix Bupleuri,Radix Glycyrrhizae.HPLC method was used for content determination of phillyrin in the preparation,the chromatographic column was Wonda Cract ODS-2 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-water(30∶70,V/V),the flow rate was 1 m L/min,the column temperature was 30℃,and the detection wavelength was 277 nm.Results The TLC spots of Flos Lonicerae,Folium Isatidis,Radix Bupleuri,Radix Glycyrrhizae were clear and the negative control showed no interference.Phillyrin showed a good linear relationship with the peak area in the range of 0.00663-0.22100 mg/m L(r=0.99999).The average recovery of phillyrin was104.45%,and the RSD was 0.79%(n=9).Conclusion The specificity,precision,stability and reproducibility of the method comply with the requirements of methodology,which can be used for quality control of Reduning Oral Liquid.
