虎黄烧伤搽剂联合硫酸庆大霉素对深二度烧伤大鼠创面愈合及免疫系统变化的影响

摘    要:目的研究虎黄烧伤搽剂联合硫酸庆大霉素对深二度烧伤大鼠创面愈合及免疫系统变化的影响。方法将SD大鼠分为对照组、模型组、硫酸庆大霉素组(模型组+硫酸庆大霉素)、虎黄烧伤搽剂(模型组+虎黄烧伤搽剂喷剂)和联合组(模型组+硫酸庆大霉素+虎黄烧伤搽剂)。给药3,14,21 d以测量卡尺检测烧伤创面愈合率;以流式细胞术检测大鼠脾淋巴液中Treg、CD4+、CD8+细胞分布。结果创伤后第21天,对照组、模型组、硫酸庆大霉素组、虎黄烧伤搽剂组、联合组的创面愈合率分别为100%,(58.29±4.39)%,(74.13±6.09)%,(76.72±5.68)%,(85.17±6.89)%。对照组、模型组、硫酸庆大霉素组、虎黄烧伤搽剂组、联合组Treg细胞比例分别为(6.57±0.18)%,(26.83±3.87)%,(19.79±2.64)%,(17.35±2.96)%,(11.03±1.52)%;CD4+分别为(79.03±5.12)%,(42.35±3.38)%,(49.17±3.08)%,(51.34±3.61)%,(63.17±3.87)%,CD8+分别为(19.32±3.38)%,(33.16±3.69)%,(27.38±3.45)%,(27.06±3.32)%,(21.05±2.49)%。各组大鼠创面愈合率随创伤时间延长而升高(均P<0.05)。与对照组相比,模型组Treg、CD8+细胞比例升高,CD4+比例降低(均P<0.05);与模型组相比,硫酸庆大霉素组、虎黄烧伤搽剂组、联合组CD4+比例升高、Treg、CD8+细胞比例降低(均P<0.05);与硫酸庆大霉素组、虎黄烧伤搽剂组相比,联合组CD4+比例升高、Treg、CD8+细胞比例降低(均P<0.05)。结论虎黄烧伤搽剂联合硫酸庆大霉素能够缓解深二度烧伤大鼠受损创面,降低机体炎症反应,其机制可能与调控脾T淋巴细胞免疫反应有关。

Abstract:Objective To explore the influences of Huhuang Burn Liniment combined with gentamicin sulfate(GS)on wound healing and immune system change in deep second degree burn rats.Methods SD rats were divided into control group,model group,gentamicin sulfate group(model+gentamicin sulfate),Huhuang Burn Liniment group(model+Huhuang Burn Liniment),combined group(model+gentamicin sulfate+Huhuang Burn Liniment).Measurin calipers on 3,14,21 d was used to detect burn wound healing rate;flow cytometry was used to detect the distribution of Treg,CD4^+,CD8^+cells in the splenic lymph fluid of rats.Results On 21 d after trauma,the wound healing rates in control group,model group,sulfuric acid gentamicin group Huhuang Burn Liniment group and combined group were 100%,(58.29±4.39)%,(74.13±6.09)%,(76.72±5.68)%,(85.17±6.89)%.The proportion of Treg cells in control group,model group,gentamicin sulfate group,Huhuang Burn Liniment group,and combined group were(6.57±0.18)%,(26.83±3.87)%,(19.79±2.64)%,(17.35±2.96)%,(11.03±1.52)%;CD4^+were(79.03±5.12)%,(42.35±3.38)%,(49.17±3.08)%,(51.34±3.61)%,(63.17±3.87)%,CD8^+were(19.32±3.38)%,(32.16±3.69)%,(27.38±3.45)%,(27.06±3.32)%,(24.05±2.49)%.The wound healing rate of rats in each group was increased with the prolongation of wound time(all P<0.05).Compared with control group,the ratio of Treg and CD8^+cells in model group were increased,and the ratio of CD4^+was decreased(P<0.05);compared with model group,the ratio of CD4^+in gentamicin sulfate group,Huhuang burn liniment group and combined group were increased,and the ratio of Treg and CD8^+cells was decreased(all P<0.05);compared with gentamicin sulfate group and Huhuang burn liniment group,the ratio of CD4^+in combined group was increased,and the ratio of Treg and CD8^+cells were decreased(all P<0.05).Conclusion The combination of Huhuang Liniment and gentamicin sulfate can relieve the damaged wound of deep second degree burn rats and reduce the inflammatory response of the body,the mechanism may be related to the regulation of the immune response of the spleen T lymphocyte.
