摘    要:目的优选地稔多糖的提取工艺。方法采用苯酚硫酸法测定多糖含量,通过单因素考察,比较微波提取与回流提取的多糖提取率;在此基础上以多糖提取得率为评价指标,从提取时间、料液比、提取次数为3因素,采用正交试验优化提取工艺。结果回流提取的多糖提取率高于微波提取;回流提取优化工艺条件为加20倍水,提取3次,每次2 h。结论回流提取方法更适用于提取地稔多糖,优化后的工艺稳定可行。

Abstract:Objective To optimize the extraction process of polysaccharide from Melastoma dodecandrum.Methods The content of polysaccharide was determined by phenol sulfuric acid method,and the effect of microwave extraction and reflux extraction on the yield of polysaccharide was compared by single factor investigation.On this basis,the extraction process was optimized by orthogonal test with the yield of polysaccharide as evaluation index,extraction time,solid-liquid ratio and extraction times as three factors.Results The yield of polysaccharide extracted by reflux extraction was higher than that extracted by microwave extraction,and the optimum conditions of reflux extraction were as follows:adding 20 times water,extracting three times,2 h each time.Conclusion Reflux extraction method is more suitable for the extraction of polysaccharide from Melastoma dodecandrum,and the optimized process is stable and feasible.
