亚硒酸钠对脑缺血再灌注损伤的保护作用及对细胞凋亡的影响

摘    要:目的:探讨亚硒酸钠对脑缺血再灌注所致脑损伤的保护作用及机理。方法:沙土鼠30只,随机分为(1)假手术组,(2)缺血再灌注组,(3)亚硒酸钠预防组。采用双侧颈总动脉夹闭法复制沙士鼠脑缺血再灌注模型,术后48h取材测定血清、脑组织匀浆中GSH-Px活力;光、电镜观察海马CA1区神经细胞形态的改变;TUNEL法显示凋亡细胞。结果:亚硒酸钠组脑组织GSH-Px活力明显增加,与缺血再灌注组比较有显著差异(P<0.05);光、电镜观察亚硒酸钠组能使脑缺血再灌注所致海马CA1区神经细胞的损伤明显减轻,细胞凋亡的表达明显减少,与缺血再灌注组比较有显著差异(P<0.001)。结论:亚硒酸钠对缺血再灌注所致的神经细胞损伤有明显的保护作用。

Abstract:Objective: To observe the preventive effect and mechanism of ample selenium on experimental cerebral ischemia reperfusion damage Methods: Thirty pallasiomies were randomly divided into three groups: (1) sham operation, (2) ischemia reperfusion and (3) ample selenium prevention Cerebral ischemia reperfusion replicas of the pallasiomies were made by clipping bilateral common carotid arteries after seven minutes in both the second and the third groups, but not in the sham operation group Forty eight hours after the operation, the pallasiomies were killed The activity of GSH Px in the brain and serum was measured Neurocyte structure in hippocapus CA 1 area was observed with optical and electron microscope respectively Apoptosis was observed by the TUNEL method Results:The activity of GSH Px in the brain and serum in the ample selenium group was high compared to that in the ischemia reperfusion group(P<0 01, P<0 05 respectively) Neurocyte structure observations showed that sodium selenite could relieve neurocyte damage caused by ischemia reperfusion in hippocapus CA 1 area The number of apoptosis was low in the ample selenium group compared to that in the ischemia reperfusion group, and there was significant difference (P<0 001) Conclusion: Sodium selenite can obviously protect neurocytes from the damage caused by ischemia reperfusion
