非酒精性脂肪肝患者肿瘤坏死因子-α基因多态性研究

目的:了解非酒精性脂肪肝(NAFLD)患者中肿瘤坏死因子 α(TNFα)启动子基因多态性在广东汉 族人群的分布及其与疾病的相关性。方法:用聚合酶链反应 限制性片段长度多态性技术检测117例NAFLD 患者和120例健康对照者外周血的TNFα基因启动子区域-308、-238位点基因多态性。结果:TNFα基因- 238位点在NAFLD组中变异的G/A基因型频率比对照组明显升高(29.9%比15.8%,P<0.05),而-308位 点无显著性差异(11.0%比8.0%P>0.05)。-238位点等位基因与NAFLD有相关性。结论:TNFα基因-238 位点G/A的突变与NAFLD易感性相关。

Tumor Necrosis Factor Alpha Promoter Polymorphisms in Non-alcoholic Fatty Liver Diseases

Objective:To explore the prevalence of the -238 and -308 TNF-alpha polymorphisms, and their relationship to non-alcoholic fatty liver diseases.Methods: The determination of TNF-alpha -238 and -308 alleles was performed in 117 patients with non-alcoholic fatty liver diseases(NAFLD) and 120 healthy controls by polymerase chain reaction and restriction fragment length polymorphism techniques. Results: The G/A mutation of the -238,but not of the -308 TNF-alpha polymorphism was higher in patients with NAFLD than in controls (29.9% vs 15.8%, P<0.05).Conclusions: There is an association between the TNF-alpha promoter polymorphism at position -238 with NAFLD.susceptibility.
