摘    要:目的:研究桑枝、桑白皮体外降糖作用,初步探讨其降糖作用机理.方法:采用与人肝细胞表型相似的HepG2细胞,检测24h后培养液中葡萄糖的消耗量,用MTT法监测细胞增殖的情况.结果:桑枝、桑白皮在高糖状态下可使HepG2细胞的葡萄糖消耗量增加,对胰岛素刺激的HepG2细胞的葡萄糖消耗量增加有协同作用.结论:促进外周组织的葡萄糖代谢、提高肝细胞对胰岛素的敏感性可能是桑枝、桑白皮防治2型糖尿病作用的机理之一.

Abstract:Objectsive:To observe the effects of extracts of Ramulus Mori and Cortex Mori on glucose uptaking in cultured HepG2 cells in vitro andinvestigate the hypoglycemic mechanism of them initially.Methods:HepG2 cells ,the phenotype of which is similar to that of the humanhepatocytes , were used to detect the 24 hours, consumption of glucose in cultured fluid , MTTassay was used to monitor the proliferation ofthe HepG2 cells.Results:Ramulus Mori and Cortex Mori could increase the glucose consumption of HepG2 cells and enhance the glucoseconsumption in coordination with insulin.Conclusions:Promoting the glucose uptake rate of peripheral tissue and enhance the sensitivity ofhepatocytes to insulin may be one of the possible hypoglycemic mechanisms of Ramulus Mori and Cortex Mori.
