苦瓜总皂苷对2型糖尿病肾病大鼠肾脏组织中单核细胞趋化蛋白1表达的影响

摘    要:目的:探讨苦瓜总皂苷对2型糖尿病肾病大鼠肾脏组织中单核细胞趋化蛋白1(MCP-1)表达的影响,分析其对2型糖尿病肾病大鼠炎症的作用。方法:将48只雄性SD大鼠随机分为空白组、模型组以及苦瓜总皂苷治疗组,每组16只。左肾切除+一次大剂量链脲佐菌素腹腔注射建立动物模型,模型建立成功后,空白组和模型组给予生理盐水灌胃,苦瓜总皂苷治疗组给予苦瓜总皂苷灌胃,分别于实验第4周、8周处死大鼠各半,测血糖、24小时尿蛋白定量以及炎性因子;免疫组化、RT-PCR以及Western Blot检测肾脏组织中MCP-1的变化特点。结果:苦瓜总皂苷治疗组尿蛋白和血清肌酐与模型组相比明显减少,血糖明显升高,差异有统计学意义(P0.05);苦瓜总皂苷治疗组与模型组相比,白介素1α(IL-1α)、白介素6(IL-6)和肿瘤坏死因子α(TNF-α)明显降低,差异有统计学意义(P0.05)免疫组化、RT-PCR和Western Blot可见:与模型组相比,治疗组大鼠肾脏组织MCP-1表达明显降低(P0.05)。结论:苦瓜总皂苷可以通过抑制糖尿病肾病大鼠肾组织MCP-1的表达,进而降低炎症反应,改善肾功能,最终调节2型糖尿病肾病的症状。

Effects of Total Momordicosides on the Expressions of MCP-1 in the Kidney of Type 2 Diabetic Nephropathy Rats

Abstract:Objective:To analyze the effects of total momordicosides against the inflammation of the rats with type two diabetic nephropathy by observing its influence on the expressions of MCP-1 in renal tissue of the rats. Methods: All 48 male SD rats were randomly divided into the blank group, the model group and the treatment group of total momordicosides,16 cases in each group. Animal models were established by left nephrectomy and intraperitoneal injection of streptozotocin in large dose for once. After the models were successfully built, the blank group and the model group were given intragastric administration of physiological saline, and the treatment group of total momordicosides accepted intragastric administration of total momordicosides. Half of the rats in each group were put to death at 4th, 8th week of the experiment respectively. Blood glucose, 24 hour urine protein quantity and inflammatory factors were tested, and the changes of MCP-1 in renal tissue were determined by immunohistoche-mistry, RT-PCR and Western Blot. Results: Urinary protein and serum creatinine decreased significantly in the treatment group of total momordicosides compared with the model group, and blood glucose increased significantly, the differences were statistically significant (P<0.05); The levels of IL-1α, IL-6 and TNF-αdecreased significantly in the treatment group of total momordicosides compared with the model group, the difference was statistically significant (P<0.05). Immunohistochemistry, RT-PCR and Western Blot results showed that the expressions of MCP-1 in rat kidney decreased significantly in the treatment group of total momordicosides compared with the model group (P<0.05). Conclusion: By restraining the expressions of MCP-1 in renal tissue of diabetic nephropathy rats, total momordicosides can decrease inflammatory response, improve kidney functions, consequently to regulate the symptoms of type 2 diabetic nephropathy.
