甘草喷雾剂对大鼠全麻气管插管后呼吸道黏膜及细胞因子的影响

摘    要:目的:探讨甘草喷雾剂对实验大鼠全麻气管插管后呼吸道黏膜组织病理学变化及血清肿瘤坏死因子α(TNF-α)、白介素1(IL-1)及白介素10(IL-10)含量的影响。方法:选择50只SPF级Wistar大鼠,雌雄各半,按体质量随机分为生理盐水组,利多卡因组,甘草喷雾高、中、低剂量组,每组10只。采用气管插管法制作动物模型。持续插管60分钟后,拔出气管导管,分别摘取各组动物咽喉部黏膜及其下组织用10%福尔马林溶液固定,切片,进行病理组织学检查。各组动物股动脉取血,离心抽取血清,检测TNF-α、IL-1及IL-10的含量。结果:大鼠血清TNF-α、IL-1、IL-10含量及炎细胞浸润程度评分甘草喷雾高、中剂量组与生理盐水组、甘草喷雾低剂量组比较,差异均有统计学意义(P0.05);甘草喷雾高、中剂量组与利多卡因组比较,差异均无统计学意义(P0.05);甘草喷雾高剂量组与甘草喷雾中剂量组比较,差异均无统计学意义(P0.05)。甘草各剂量组黏膜上皮表面炎性渗出均较生理盐水组减轻,固有膜内炎细胞浸润均较生理盐水组减轻。结论:甘草喷雾剂对气管插管后呼吸道黏膜具有保护作用。

Abstract:Objective:To study the effects of licorice spray on pathological changes of respiratory mucosa and content of TNF-α, IL-1 and IL-10 in rats which were given general anesthesia tracheal intubation. Methods:Fifty SPF Wistar rats in half genders were selected and randomly divided into normal saline group, lidocaine group and low, middle and high dose groups of licorice spray according to the weight, each group 10 rats. Tracheal intubation was adopted to establish animal models. Tracheal tube was pulled out after 60 minutes of continuous intubation, and then the mucous membranes of the throat and tissues from each animal were obtained, fixed in 10%formalin solu-tion, sectioned and histopathology examinations were performed. The blood was drawn from the femoral artery, cen-trifugal extraction of serum was made, the contents of TNF-α, IL-1 and IL-10 were tested. Results:The score of con-tent of TNF-α, IL-1 and IL-10 and the degree of infiltration of inflammatory cells were compared between high and middle dose groups of licorice spray with normal saline group and low dose group of licorice spray, the differences were statistically significant (P< 0.05). The differences between high and middle dose groups licorice spray and lidocaine group were not significant (P>0.05). There was no significant differences between high and middle dose groups of licorice spray(P>0.05). Inflammatory exudate in the surface of epithelium mucosa and intrinsic membrane inflam-matory cells infiltration in all dosage licorice spray groups were less than that in saline group. Conclusion:Licorice spray has protective effects on mucous membrane of respiratory tract after tracheal intubation.
