骨髓增生异常综合征患者血小板内环核苷酸的变化

我们用放射免疫分析测定了２５例骨髓增生异常综合征（ＭＤＳ）、１８例急性非淋巴细胞性白血病（ＡＮＬＬ）、２９例正常人血小板内环一磷酸腺苷（ｃＡＭＰ）和环一磷酸鸟苷（ｃＧＭＰ）的含量，ＭＤＳ和ＡＮＬＬ病人血小板内ｃＡＭＰ和ｃＧＭＰ含量均明显高于正常对照（Ｐ均＜０．０１），而ＡＮＬＬ病人血小板内ｃＡＭＰ又高于ＭＤＳ病人（Ｐ＜０．０１），血小板内ｃＡＭＰ／ｃＧＭＰ（ｃＡ／ｃＧ）比值则缺乏规律性变化。ＭＤＳ病人的血小板聚集明显低于正常对照（Ｐ＜０．０１）．两组病人外周血小板数明显低于正常（Ｐ＜０．０１），且在正常组中，血小板内ｃＡＭＰ含量与血小板数成负相关（ｒ＝－０．７０６９，Ｐ＜０．０１）。结果提示，ＭＤＳ病人血小板聚集功能减退可能与血小板内ｃＡＭＰ和ｃＧＭＰ含量升高有关，且血小板内ｃＡＭＰ含量随病情的恶性程度增加而升高。

Platelet Cyclic Nucleotide Levels in Patients With Myelodysplastic Syndrome

Platelet cyclic AMP (cAMP) and cyclic GMP (cGMP) levels were measured by 125IcAMP and 125I-cGMP RIA in 25 patients with myelodysplastic syndrome (MDS), 18 cases of acute nonlymphocytic leukemia (ANLL) and 29 healthy subjects. The platelet cAMP and cGMP levels in patients with MDS or ANLL were significanly higher than those in the normal subjects (P<0. 01,respectively). The platelet cAMP levels of the patients with ANLL were significantly elevated as compared with those of the patients with MDS (P<0. 01). In contrast, no consistent change was found in platelet cAMP/cGMP ratio. Platelet aggregation induced by ADP was significantly reduced in 6 patients with MDS than in 4 normal subjects (P<0.01 ). The mean platelets count was reduced in both patients with MDS and ANLL compared to that of normal subjects (P<0. 01, respectively), and in normal subjects, the platelet cAMP level was inversely correlated with the platelet count (r=-0. 7069, P<0. 01). The results suggest that the platelet aggregation disorder is related to the elevation of the platelet cAMP and cGMP levels in patients with MDS,and the platelet cAMP levels is increased with the malignancy of the disease.
