摘    要:应用三种培养方法观察人角化细胞生长、增殖与形成膜片的过程,共111次。人表皮细胞单层细胞生长率3T3饲养层培养法最高达80.0%;皮块培养法为52.5%;细胞悬液培养法仅为11.8%。3T3饲养层培养法效果最佳,优点较多,值得选用。皮块培养法简便易行,效果也可,如能设法排除成纤维细胞混合生长,仍为可选用的培养方法。

Abstract:In order to select a more efficient method for cultivation of human epidermal keratinocytes in the treatment of major burns, three kinds of culture models were established.The growth of 111 cultures of human epidermal keratinocyes in explant culture,dispersed cell culture and 3T3 feeder layer culture were studied.80% of cultures were confluent in 3T3 feeder layer culture,52.5% of cultures in explant culture and 11.8% of cultures in dispersed cell culture. The pattern of growth of cultured keratinocytes in each model were described.lt is concluded that among the models studied, the 3T3 feeder layer culture is the most effective and posseses many advss tages for cultivation of human keratinocytes;the explant culture would be a simpls aad easy method if one can find out an effective means of controling fibroblasts con tamination.
