人骨巨细胞瘤细胞DNA含量的研究

摘    要:将体外培养的人骨巨细胞瘤第四代和第五代瘤细胞,进行~3H—TdR放射自显影术和Feulgen反应,先计算出银粒标记细胞(S,G_2)的增殖比率,再用显微分光光度法测定未被标记的细胞(G_0和G_1期)的DNA含量。结果,第四代单核瘤细胞DNA含量为亚四倍体值((?)=87.36,DI=1.83),第五代单核瘤细胞DNA含量为超四倍体值((?)=118.13,DI=2.47).两代瘤细胞DNA含量经统计学处理,有显著差异(P<0.01),证实了骨巨细胞瘤细胞长期传代培养可以恶化。

Abstract:The cells of human giant cell tumor of bone obtained from surgical clinics were cultured in vitro according to the routine procedure. Mononuclear tumor cells grown from the explants were analyzed by 3H-TdR autoradiography and cytophotome-try. The proportion of proliferative cells (S, G2 ) labeled by silver grains was counted, then Feulgen-density of unlabeled cells(G0, G1)in the same slide was determined microspectrophotometrically. Only a small number of tumor cells were labeled by silver grains. The DNA content (optical density = 47.75) of lymphocytes in human peripheral blood is used as reference (diploid value) standard, DNA indices ( DI ) were calculated from the ratio of the DNA content in analyzed cells to normal diploid value. The cellular DNA content in tumor cells of the fourth generation is hypotetraploid (mean 87.34, DI=1.83) , however, in tumor cells of the fifth generation it is hypertetraploid ( mean 118.13, DI = 2.474) .The difference of DNA values between the two generations is statistically very significant(P<0. 01). The results support the view that mononuclear stromal cells in giant cell tumor of bone may change into more malignant from in vitro, and multinuclear giant cells may be benign in nature.
