胃癌病理类型及分期与胃癌组织中PI3K/CREB表达水平的相关性分析

目的：探究胃癌病理类型及分期与胃癌组织中磷脂酰肌醇3- 激酶（PI3K）/cAMP 反应序列结合蛋白（CREB）表达水平的相关性。方法：选取140 例在本院手术的胃癌患者病例标本作为检测样本，同时选取胃癌患者距肿瘤边缘1 cm 处的癌旁组织标本作对比。用免疫组织化学、免疫印迹检测样本中PI3K、CREB 阳性率及蛋白表达量。结果：PI3K、CREB 在胃癌组织中的阳性率高于癌旁组织；PI3K/CREB 在胃癌组织中的蛋白表达量均高于癌旁组织；胃癌组织中PI3K、CREB 在低分化、Ⅲ+Ⅳ期中阳性表达率及蛋白水平均高于高中分化、Ⅰ+Ⅱ期中的表达；PI3K、CREB 阳性胃癌患者生存率低于阴性患者；PI3K 诊断胃癌的ROC曲线下面积（AUC）为0.765，灵敏度为78.64%、特异度为62.46%，CREB 诊断胃癌AUC 面积为0.774，灵敏度为70.93%、特异度为64.75%。结论：PI3K、CREB 在胃癌组织中呈现高表达，且与胃癌疾病进展存在相关性，对胃癌预后诊断具有重要意义。

Correlation between pathological types and stages of gastric cancer and PI3K/CREB expression in gastric cancer tissues

Objective To explore the correlation between the pathological types and stages of gastric cancer and theexpression of phosphatidylinositol 3-kinase（PI3K）/responsive element binding protein（CREB） in gastric cancertissues. Methods One hundred and forty surgical specimens of gastric cancer patients were selected from our hospitalas test samples. At the same time， a sample of adjacent tissues fromgastric cancer patients 1 cm from the edge of thetumor was selected for comparison. PI3K，CREB positive rate, and protein expression samples were detected byimmunohistochemistry and Western blotting. Results The positive rate of PI3K and CREB in gastric cancer tissueswas higher than that in adjacent tissues. The protein expression of PI3K/CREB in gastric cancer tissues was higherthan that in adjacent tissues. The positive expression rate and protein level of PI3K and CREB in poorly differentiatedand stage Ⅲ+Ⅳ gastric cancer tissues were higher than those in high-medium differentiation and stage Ⅰ+Ⅱ . Thesurvival rate of PI3K and CREB positive gastric cancer patients was lower than that of negative patients. PI3Kdiagnoses gastric cancer showed an AUC area of 0.765， a sensitivity of 78.64%， and a specificity of 62.46%. TheAUC area of CREB in the diagnosis of gastric cancer was 0.774， the sensitivity was 70.93%， and the specificitywas 64.75%. Conclusion PI3K and CREB are overexpressed in gastric cancer tissues and are correlated with theprogression of gastric cancer， which is of great significance for the prognostic diagnosis of gastric cancer.
