当归红芪超滤膜提取物含药血清对辐射心肌成纤维细胞SOD和羟脯氨酸的影响

摘    要:目的观察当归红芪超滤膜提取物含药血清对辐射心肌成纤维细胞SOD和羟脯氨酸的影响。方法用胰酶-胶原酶消化法分离培养乳鼠心肌成纤维细胞,建立体外培养体系,通过10万分子量的超滤膜提取物制备不同浓度和不同期限的含药血清。将已经过药物干预24 h的第2代细胞进行辐射剂量为10 Gy的辐射,继续培养24 h,黄嘌呤氧化法检测超氧化物歧化酶(SOD)活性,碱水解法检测羟脯氨酸含量。结果与空白组比较,模型组SOD活性显著下降,羟脯氨酸含量显著增加(P0.01);与模型组比较,药物组SOD活性显著升高(P0.05或P0.01),且药物组(7 d)显著优于药物组(3 d)(P0.05),而羟脯氨酸含量下降并不显著(P0.05)。结论辐射可引起SOD活性下降、羟脯氨酸含量增加,而当归红芪超滤膜提取物能去除氧自由基,并且长期疗效优于短期,但对羟脯氨酸含量并无影响。

Effects of serum containing gradocol membranes extract from Angelica Sinensis and Radix Hedysari on superoxide dismutase and oxyproline hydroxyproline of radiation cardiac fibroblasts

Abstract:Objective To observe the effects of serum containing gradocol membranes extract from Angelica Sinensis(AS) and Radix Hedysari (RH) on superoxide dismutase (SOD) and oxyproline hydroxyproline (Hyp) of radiation cardiac fibroblasts. Methods Cardiac fibroblasts (CFs) of neonatal rats were isolated and cultured by digestive method of pancreatin and collagenase, and the cell culture systems were established in vitro. Production of different concentrations and different time of drug-containing serum was be obtained by gradocol membranes extract from AS and RH with 100 000 molecular weight. The second passages of cardiac fibroblasts that were intervened by drug for 24 hours were given radiation of 10 Gy X-ray, and continued to be cultured for 24 hours, and measured SOD and Hyp by xanthine oxidase method and basic hydrolysis method. Results Compared with blank group, the activity of SOD of model group was obviously decreased, and the content of Hyp was obviously increased(P 〈0.01 ). Compared with model group, the activity of SOD of drug group was obviously increased(P 〈 0.05 or P 〈 0.01 ) ,and drug group of 7 days was better than 3 days (P 〈 0.05 ) , but the content of Hyp wash' t obviously decreased (P 〉 0.05). Conclusion The radiation can cause the activity of SOD to decrease and the content of Hyp to increase. Gradoeol membranes extract from AS and RH can remove oxygen free radicals, and the long-term efficacy was better than the short-term, but it isn' t effect on the content of Hyp.
