大鼠心肌梗死后血浆Ghrelin的动态变化

摘    要:目的探讨大鼠心肌梗死后血浆Ghrelin水平改变及与心功能改变的关系。方法30只Wistar大鼠被随机分成对照组(n=6只),假手术组(n=6只),心肌梗死3d组(n=6只),梗死7d组(n=6只)和梗死28d组(n=6只)。通过结扎冠状动脉前降支制作心肌梗死动物模型,假手术组开胸后仅在冠状动脉下穿线,但不结扎。血流动力学检查记录心率(HR)、左心室收缩压(LVSP)、左心室舒张末期压力(LVEDP)以及压力上升最大速率(dp/dtmax)。对各组动物采集外周血并制备血清,采用ELISA方法检测血浆Ghrelin浓度。结果与假手术组相比,梗死组大鼠LVSP和dp/dtmax显著降低(P〈0.05),而LVEDP明显增高(P〈0.05)。与梗死后3d及7d组比较,梗死28d组LVEDP明显增高(P〈0.05)。心肌梗死后大鼠不同时间点Ghrelin血浆浓度逐渐降低(P〈0.05),而假手术组与对照组相比较,血浆Ghrelin浓度差异无统计学意义(P〉0.05)。结论心肌梗死后大鼠血浆Ghrelin浓度改变可能与梗死后心脏重构相关。

Abstract:Objective To observe the correlation between cardiac function and the dynamic expression of Ghrelin from 3 days to 28 days in rats with myocardial infarction. Methods 30 Wistar rats were divided into four groups by the way of random number. They were control group (n=6), sham-operated group (n=6), MI-3 d group (n=6), MI-7 d group (n=6) and MI-28 d group (n=6). The left anterior descending (LAD) was ligated and the sham animals underwent thoracotomy and pericardiotomy, but not LAD ligation. All the animals in each groups were underwent cardiac catheterization, include heart rate, left ventricular systolic pressure, left ventricle end-diastolic pressure and dp/dtmax. The concentration of Ghrelin in plasma was measured by using enzyme-linked immunosorbent assay (ELISA). Results Compared with control group and sham-operated group, the LVSP and dp/dtmax at each MI-groups was markedly decreased (P 〈 0.05), but LVEDP was increased (P 〈 0.05). compared with the MI-3 d and MI-7 d group, LVEDP was increased in MI-28 d group (P 〈 0.05). The expression of Ghrelin was dy- namic reduction in rats with myocardium infarction (P 〈 0.05). There was no different of Ghrelin level in sham-operated group and control group. Conclusion This change of Ghrelin expression may be related to cardiac remodeling after myocardium infarction.
