| Abstract: | Viable cell analysis and prenatal chromosomal diagnosis with improved technique of amniotic fluid cell culture were performed in 105 cases. The procedures of media renewing were modified, harvest criteria were established and methods of harvesting culture were improved. Various patterns of chromosome banding were selectively used. By means of the above improved techniques, successful culture rate approached 99.05% and duration of cultivation shortened to an average of 7.6 days. Four amniotic fluid cell cultures contaminated by RBC more than 10~7/ml yielded satisfactory results. Eleven pregnant women at high risk of having chromosome defective babies were diagnosed and underwent abortion far eugenics. |
