缺氧诱导因子-1α和转化生长因子-β3在子痫前期患者胎盘组织中的表达

摘    要:目的:研究缺氧诱导因子-1α(HIF-1α)和转化生长因子-β3(TGF-β3)在子痫前期患者胎盘组织中的表达及意义。方法:采用免疫组化方法和图像分析系统分别对20例正常孕妇(对照组)和46例子痫前期患者(子痫前期组,其中轻度16例、重度30例)胎盘组织中的HlF-1α和TGF-β3的表达进行分析。结果:①HIF-1α主要表达于胎盘绒毛滋养细胞和蜕膜细胞,与正常组胎盘组织(65.2±1.77)比较,子痫前期组HIF-1α的平均光密度(78.10±3.84)显著升高(P<0.05),重度子痫前期(81.49±1.81)显著高于轻度子痫前期(74.71±1.53)(P<0.05)。②TGF-β3主要表达于胎盘绒毛滋养细胞和蜕膜细胞,与正常组胎盘组织(63.55±2.10)比较,子痫前期组TGF-β3的平均光密度(75.34±3.73)显著升高,其中重度子痫前期(78.05±3.14)显著高于轻度子痫前期(72.64±1.79)。③子痫前期组HIF-1α和TGF-β3的表达呈正相关性(γ=0.351,P<0.05)。其中轻度子痫前期γ1=0.549,P<0.05。重度子痫前期γ2=0.577,P<0.05。结论:胎盘滋养细胞HIF-1α和TGF-β3表达升高可能与子痫前期的发病有关。

Expression of hypoxia - inducible factor - 1α and transforming growth fator - beta3 in the placentas of pre-eclampsia
Abstract:Objective:To discuss the expression of hypoxia-inducible factor-lα and transforming growth fator-beta3 in the placentas of pre-eclampsia.Methods:Immunohistochemical technique and image analysis system were applied to detect and analyze the expression of HIF-lα and TGF-β3 in the placentas from pre-eclampsia(n=46,16 women of mild pre-eclampsia and 30 women of severe pre-eclampsia)and normotensives(n=20).Results:(1)HIF-lα was mostly found in trophoblast and decidual cells.The mean density of pre-eclampsia placentas(78.10±3.84)was significantly higher than that of normotensives(65.24±1.77)(P< 0.05).The mean density of severe pre-eclampsia placentas(81.49±1.81)was significantly higher than that of mild pre-eclampsia(74.71±1.53)(P< 0.05).(2)TGF-β3 was also mostly found in trophoblast and decidual cells.The mean density of pre-eclampsia placentas(75.34±3.73)was significantly higher than that of normotensives(63.55±2.10).The mean density of severe pre-eclampsia placentas(78.05±3.14)was significantly higher than that of mild pre-eclampsia(72.64±1.79)(P<0.05).(3)There was positive correlation between the expression of HIF-lα and TGF-β3 in mild and severe pre-eclampsia(γ_1=0.549,γ_2=0.577,P<0.05).Conclusion:HIF-lα and TGF-β3 in human placentas are mainly located in trophoblast and decidual cells.Increasing HIF-lαand TGF-β3 expression may have an important role in the pathogenesis of pre-eclampsia.
