SELDI-TOF-MS技术结合LCM技术分析结肠癌细胞蛋白质组学构成

目的表面增强激光解吸电离质谱(SELDI-TOF-MS)技术结合激光捕获微切割(LCM)分析正常结肠粘膜上皮细胞和结肠癌细胞的蛋白质组学构成.方法将新鲜结肠标本常规制备冰冻组织切片,通过激光捕获微切割从组织冰冻切片中分别切取结肠粘膜上皮细胞和结肠癌细胞,SELDI技术分析蛋白质组学构成.结果激光捕获微切割可正确有效地分离结肠癌和正常粘膜上皮细胞;比较结肠癌和正常结肠粘膜细胞的飞行质谱图提示有多处峰存在显著性差异.结论 SELDI技术结合激光捕获微切割技术可以快速有效分析结肠癌细胞的蛋白质构成;所获结果为结肠癌发生发展过程细胞蛋白质文库的建立提供了依据.

Analysis of colorectal cancer cell proteomics by SELDI-TOF-MS coupled with LCM

Objective To apply the surface-enhanced laser desorption/ionization(SELDI) technique coupled with laser capture microdissected(LCM) technique to analyse proteomics of human colorectal cancer cells and normal mucosa cells effectively.Methods LCM technique was utilized to obtain the cells of cancer and normal mucosa from frozen sections.SELDI-TOF-MS was used to generate protein profiles in the cells of cancer and normal mucosa.Results Some peaks were identified that significantly differed from intensities between colorectal carcinoma and adjacent normal mucosa.In addition,results of SELDI-TOF-MS were presented that several apparently distinct protein peaks of human colorectal cancers showed markedly increased compared with normal cells.Conclusion This study demonstrates that it is rapid and effective that using SELDI-TOF-MS technique to analyze distinct proteins of colorectal carcinoma cells was simple and highly reproducible.The results may provide an important reference to set up protein profiles of colorectal cancer cells.
