胃癌组织中p33~(ING1)、p53基因蛋白的表达及其临床意义

目的　探讨 p33ING1基因蛋白在胃癌中表达的临床意义及其相互关系。方法　应用Envision免疫组化法检测71例胃癌、12例胃粘膜不典型增生患者p33ING1、p5 3基因蛋白的表达 ,并与正常胃粘膜及慢性胃炎粘膜 18例对照。结果　粘膜不典型增生组及对照组中 p33ING1均阳性表达 (10 0 % ) ,而胃癌中表达 6 2 .0 % (4 4/ 71) ,明显下降 ,与前两组之间差异显著 (P <0 .0 1) ;胃癌中 p33ING1表达与肿瘤的浸润、淋巴结的转移及分化有关 (P <0 .0 1) ;p5 3表达与肿瘤大小、浸润、淋巴结转移有关 (P <0 .0 1)。结论　p33ING1是抑癌基因 ,在胃癌中表达下降 ,对胃癌发生、发展可能起重要作用 ,p33ING1和 p5 3在胃癌中表达有相关性 (P <0 .0 5 )。p33ING1与 p5 3野生型基因互相协同、依赖 ,抑制细胞生长 ,促进凋亡。检测胃癌患者p33ING1的水平 ,对p5 3介导的基因治疗有重要意义。

The Expression of Gene ING1 and p53 in Stomach Cancer and Its Clinical Signification

Objective To explore the clinical pathological significanc and relationships between p33 ING1 expression and carcinoma of stomach.Method Using Envision immunohistochemical method, the expression level of p33 ING1 and p53 was examined in 71 cases of stomach cancer, 12 cases of gastric mucous membrane atypical hyperplasia and 18 cases of control group.Result p33 ING1 expression in mucous membrane atypical hyperplasia group and control group is positive(100%), the expression in gastric carcinoma group was obviously lower(62.0%,44/71)than that in the other two groups significantly( P <0.01). p33 ING1 expression in stomach cancer was related to the tumor grouth, lymphnode metastasis and tumor polarization ( P <0.01), p53 expression was related to tumor size, growth and lymphnode metastasis ( P< 0.01).Conclusion p33 ING1 is a tumor supressor gene, its expression in carcinoma of stomach was low,it may play an important role in gastric carcinoma occurrence and development. The expression is related to the growth of gastric carcinoma, metastasis and differentiation( P <0.01). p33 ING1 is related to p53 expression in carcinoma of stomach ( P< 0.05).p33 ING1 combined with p53 wild type suppresses cell growth and accelerates cell apoptosis.p33 ING1 level is very important to gene therapy mediated by p53 and worthes studying further.
