维生素C和维生素E对移植肺再灌注损伤保护作用的实验研究

摘    要:目的:观察维生素C(Vit)和维生素E(VitE)对大鼠移植肺再灌注损伤的保护作用。方法:采用大鼠自体左肺模拟原位移植模型,治疗组分别于再灌注前10min静脉注入VitC(200mg/kg)和(或)VitE乳剂(100mg/kg)。再灌注4h后观测移植肺气体交换功能,顺应性,血管通透性,脂质过氧化程度,ATP含量以及组织病理学变化。结果:缺血再灌注(I/R)组移植肺气体交换功能,顺应性降低,脂质过氧化程度增加,ATP含量下降,组织病理学改变明显(P<0.01);给药组移植肺气体交换功能,顺应性与I/R组相比无明显改善(P>0.05),其它指标均有不同程度的改善,以VitC和VitE合用时改善最为显著(P<0.01)。结论:VitC和VitE对移植肺再灌注损伤具有协同保护作用。

Abstract:Objective The effects of vitamin C(VitC)and vitamin E(VitE)on reperfusion injury of transplanted lung in rats were observed.Methods Adult Sprague-Dawley rats were divided into five experimental groups(n=10,each group):group A,Sham-operated;group B,Ischemia-reperfusion;group C, treatment with VitC;group D,treatment with VitE;and group E,treatment with VitC plus VitE. A simulated model of orthotopic left lung autotransplantation in rats was used. VitC(200 mg/kg) and hours (or) VitE(100 mg/kg) were given through jugular veins at 10 minutes before reperfusion in treatment groups.Left lung injury was assessed at 4 hours after reperfusion by pulmonary function(using PaO 2,PaCO 2,pressure of airway),vascular permeability(using BALF albumin and water content of left lung).Malonyl dialdehyde(MDA)content,superoxide dismutase(SOD)activity,adenosine triphosphate(ATP)content,the histopathologic alternations of transplanted lungs were also observed.Results All of the indexes became worse in group B than in group A( P <0 01).For PaO 2,PaCO 2 and pressure of airway,no significant difference was found between group B and group C,group D or group E( P >0 05).But vascular permeability,lipid peroxidation,ATP content and histopathologic alternations of transplanted lung were improved to some extent in group C,D and E.And the difference was the most significant between group B and group E( P <0 01).Conclusion The VitC and VitE provided synergetically protective effects on reperfusion injury of transplanted lung.
