摘    要:目的 探讨黏着斑蛋白( vinculin,VCL)在诊断前列腺癌及判断恶性程度的作用.方法 应用基因芯片筛选出在前列腺癌与前列腺增生组织特异性表达的基因VCL,免疫组化技术检测VCL蛋白在前列腺癌与癌旁组织中的表达,结合VCL免疫组化评分和前列腺癌患者的临床病理参数进行分析.结果 通过免疫组化染色,我们发现VCL在癌旁组织中的表达明显强于前列腺癌组织(P=0.002),在前列腺癌并转移组织中的表达亦明显强于原位前列腺癌组织(P =0.003).VCL表达与前列腺癌患者血清PSA水平(P=0.011)、Gleason score(P =0.003)和肿瘤TNM分期(P=0.014)负相关,与前列腺癌转移(P=0.000)正相关,而与患者年龄(P=0.685)无明显相关.结论 VCL低表达与肿瘤的恶性程度相关,VCL缺失可能导致恶性肿瘤细胞迅速生长,可结合血清PSA水平对前列腺癌疑似患者进行早期诊断及初步判断肿瘤恶性程度.

Abstract:【Objective】 To explore the role of vinculin(VCL) in diagnosing prostate cancer(PCa) and judging the degree of the malignancy of PCa.【Method】 Using the gene chip to screen VCL in PCa and benign prostatic hyperplasia(BPH) tissues,analyzing the differentially expressed degree of VCL protein in PCa and adjacent tissues by immunohistochemical(IHC) technology,the combination of VCL IHC score and clinicopathological parameters of PCa patients were analyzed.【Results】 VCL in adjacent tissues was significantly higher than that in PCa tissues(P =0.002),and relative to primary PCa tissues,the same phenomenon was observed in PCa metastasis tissues(P =0.003).The expression of VCL was negative correlation with serum PSA levels(P =0.011),Gleason score(P =0.003) and TNM stage(P =0.014),positive correlation with metastasis(P =0.000),but not age(P =0.685).【Conclusions】 The low expression of VCL is closely related with the malignancy of PCa,and the loss of VCL leads to promote tumor cell growth,it may be combined with serum PSA levels and VCL levels to diagnosis PCa early and judge the degree of the malignancy of PCa.
