人参总皂苷抑制钙调神经磷酸酶信号通路参与其抗血管平滑肌细胞增殖作用

摘    要:目的 我们先前报道人参总皂苷(total ginsenosides,TG)具有抗血管平滑肌细胞(VSMC)增殖作用,本工作重点探讨该作用是否与其抑制钙调神经磷酸酶( calcineurin,CaN)信号通路有关.方法 采用来自大鼠胸主动脉的培养VSMCs,以real time RT- PCR检测CaN的mRNA表达,并用Western blotting检测CaN的催化亚基CnA的蛋白表达.结果 TG在抑制VSMC增殖的浓度(0.05 mg/mL和0.1 mg/mL)能显著抑制AngⅡ所引起的CaN mRNA和CnA蛋白表达的升高.结论 TG抑制CaN信号通路可能是其抗VSMC增殖的分子机制之一.

Abstract:Objective We have previously reported that total ginsenosides(TG) inhibit the proliferation of vascular smooth muscle cells(VSMCs) induced by angiotensinⅡ(AngⅡ).In the present study,it is investigated whether the inhibition of calcineurin(CaN) pathway is involved in the anti-proliferative effect of TG on VSMCs.Methods The cultured VSMCs from the rat aorta were used in the assays,and the real time RT-PCR and Western blotting were used to detect the expressions of CaN mRNA and the protein of CnA,the catalytic subunit of CaN,respectively.Results TG could remarkably inhibit the AngⅡ-induced over expressions of CaN mRNA and CnA protein at the concentrations(0.05mg/mL and 0.1 mg/mL) which could inhibit VSMC proliferation.Conclusion Inhibition of calcineurin pathway is involved in anti-proliferative effect of total ginsenosides on vascular smooth muscle cells.
