广州地区成年女性EB病毒抗体水平相关性及其年龄分布

摘    要:目的调查广州地区成年女性血清免疫球蛋白水平和抗EB病毒VCA/EBNA1 IgA和IgG抗体的年龄分布及其相互关系。方法收集广州市309例常规体检的健康女性血清样本,分别采用比浊法和ELISA检测血清免疫球蛋白水平、抗EB病毒VCA/EBNA1 IgA和IgG抗体水平。结果广州地区成年女性抗EB病毒VCA-IgA、抗VCA-IgG及抗EBNA1-IgA、抗EBNA1-IgG的阳性率分别为14.6%、92.2%、3.2%、99.7%。抗VCA-IgA阳性率与年龄呈正相关(r=0.163,P=0.004);抗EBNA1-IgG OD值与年龄呈负相关(r=-0.178,P=0.020)。各抗体水平间的相关性分析显示,抗EBNA1-IgA与血清总IgG水平之间呈负相关(r=-0.154,P=0.008);抗VCA-IgA与抗EBNA1-IgA、抗EBNA1-IgA与抗EBNA1-IgG、血清总IgA与抗VCA-IgA、总IgG与抗VCA-IgG、总IgA与总IgG、总IgM与总IgG水平之间均存在正相关(r分别为0.113、0.241、0.228、0.228、0.433和0.362,P0.05)。结论广州地区成年女性99.7%存在EB病毒既往感染。随着年龄增长,抗VCA-IgA阳性率逐渐增高,而抗EBNA1-IgG OD值则逐渐下降。抗EB病毒抗体水平间存在一定的相关性,有助于鼻咽癌等疾病筛检时联合指标的选择。

Abstract:Objective To investigate the age distribution of total immunoglobulin, IgA and IgG against Epstein-Barr virus viral capsid antigen(VCA) and nuclear antigen-1(EBNA-1) in serum among women in Guangzhou and analyze the correlation with each other. Methods Women who attended a health check-up were consecutively recruited to this study. IgA antibodies against EBV VCA and EBNA-1 were examined using ELISA and total IgA, IgM, and IgG were measured by turbidimetry. Correlations between different antibodies and age and each other were explored using Rank correlation analysis.Results A total of 309 women were tested and the positive rates were 14.6%, 92.2%, 3.2%, and 99.7% for VCA-IgA, VCAIgG, EBNA1-IgA, and EBNA1-IgG, respectively. The positive rate of EBV VCA-IgA was positively correlated with age(r = 0.163, P = 0.004); the level of EBV EBNA1-IgG was negatively correlated with age(r =-0.178, P = 0.020). Moreover,the level of total IgG was negatively correlated with the level of EBNA1-IgA; EBNA1-IgA was positively correlated with EBV VCA-IgA and EBNA1-IgG; total IgA was positively correlated with total IgG and VCA-IgA; total IgG was positively correlated with VCA-IgG and total IgM. Conclusion The infection rate of EBV was as high as 99.7% of women in Guangzhou. Along with age, the positive rate of VCA-IgA gradually increased, while the OD level of EBNA1-IgG decreased. In addition, there was a correlation among different EBV antibody levels, which would be beneficial to biomarker selection for nasopharyngeal cancer and other diseases.
