依那普利对小鼠心肌梗死后Th1/Th2、Treg/Th17的调节作用

摘    要:目的观察血管紧张素转换酶抑制剂(ACEI)依那普利对新方法诱导小鼠心肌梗死4周后Th1/Th2、Treg/Th17的平衡影响。方法 37只雄性C57BL/6小鼠用新方法结扎左冠状动脉建立心肌梗死模型(n=32),假手术组(n=5)不进行最后的冠状动脉结扎。术后将模型组存活小鼠(n=25)随机分为依那普利干预组(n=12)和心肌梗死组(n=13),依那普利干预组每日给予依那普利7.5 mg/kg灌胃,心肌梗死组和假手术组给予生理盐水灌胃。给药28 d后检查各组小鼠心脏彩超,称量小鼠心脏重量和体重,心脏采血并提取外周血淋巴细胞,采用流式细胞术检测外周血Th1和Th2、Treg和Th17细胞比率。结果依那普利干预组小鼠心脏质量/体质量比、左室舒张末内径(LVEDd)、左室收缩末内径(LVESd)均小于心肌梗死组,且均大于假手术组(P0.05);左室短轴缩短分数(LVFS)、左室射血分数(LVEF)高于心肌梗死组,且小于假手术组(P0.05)。依那普利干预组小鼠外周血Th1、Th17细胞比率明显低于心肌梗死组,高于假手术组(P0.05);而Th2、Treg细胞比率高于心肌梗死组,低于假手术组(P0.05)。结论小鼠心肌梗死4周后外周血有Th1/Th2、Treg/Th17的失衡,依那普利可以调节Th1/Th2、Treg/Th17平衡,这可能与其抑制心梗后小鼠心室重构有关。

Abstract:Objective The purpose of this study was to investigate the effect of enalapril on Th1 / Th2, Treg / Th17 of post myocardial infraction in mice at 28 days. Methods 37 male C57 / B6 mice were grouped into 2 groups at random. One group was established myocardial infarction(n=32) by ligating left anterior descending coronary artery. After operation, 25 mice survived and were further divided into 2 groups: Myocardial infarction group(MI, n=13) and Enalapril intervention group(Ena, n=12) which administration enalapril by gavage once a day with a dose of 7.5 mg / kg for 28 days. Another group of mice was the sham group(n=5). Echocardiography was performed to evaluate cardiac functions of all groups. Heart weight and boby weight of all the mice were measured at 28 days. Lymphatic cells were extracted from blood collected by heart puncture and the ratios of Th1 / Th2 and Th17 / Treg cells were measured by flow cytometry. Results Compared with MI group the ratio of heart weight, LVEDd and LVESd in Ena group was lower and the percentage of Th1, Th17 cell were significantly downregulated(P〈0.05), whereas the percentage of Th2, Treg, LVFS and LVEF were significantly increased(P〈0.05). The ratio of heart weight, LVEDd and LVESd in Ena group was higher and the percentage of Th1, Th17 cell were significantly upregulated when compared with Sham group(P〈0.05), while the percentage of Th2, Treg, LVFS and LVEF were significantly decreased(P〈0.05). Conclusion Th1 / Th2, Th17 / Treg imbalance occurred in mice with post myocardial infraction, Enalapril can regulate Th1 / Th2, Th17 / Treg imbalance which may have association with the effect of inhibit ventricular remodeling.
