中重度哮喘急性发作患儿血中EAN-78的表达及甲基强的松龙对其表达的影响

摘    要:目的:观察中重度哮喘急性发作患儿血中内皮源性中性粒细胞激活肽-78(EAN-78)的表达水平及甲基强的松龙(甲强龙)对其的影响。方法:选择中重度哮喘急性发作患儿为观察对象,分为甲强龙干预前、后组,小儿外科斜疝患儿(无哮喘病史)为对照组,留取外周血标本检测中性粒细胞(PMN)数目,并采用流式细胞术检测EAN-78的含量。结果:观察组血ENA-78水平明显高于对照组(P〈0.01),其中干预前组血ENA-78水平又明显高于干预后组(P〈0.01);观察组外周血PMN均分别较对照组明显增高,差异有统计学意义(P〈0.01),但干预前、后两组外周血PNN计数比较差异无统计学意义(P〉0.05);甲强龙干预前组血ENA-78的表达与PNN数目呈显著正相关(n=32,r=0.865,P〈0.01)。结论:中重度哮喘急性发作时患儿血中ENA-78的表达及PMN数量是升高的,它们可能共同参与了哮喘发病的炎症过程;甲强龙部分通过抑制ENA-78的表达来减轻气道炎症。

Abstract:AIM: To observe the expressions of epithelial cell-derived neutrophil-activating protein-78 in acute asthma of severe cases and the influence of methylprednisolone. METHODS: Children with moderate and severe acute asthma were divided into urbason pre-interfere group, post-interfere group, and the pedi-atric surgery indirect hernia children as control group (without asthma case history) , the numbers of hetero-phil granulocyte ( PMN ) in peripheral blood preparation were detected, and the expressions of EAN-78 protein were measured by flow cytometry. RESULTS: Compared with the control group, the levels of ENA-78 protein in asthma group were higher( P < 0.01 ). Compared with post-interfere group, the levels of ENA-78 protein in pre-interfere group were significantly higher (P < 0.01). Compare with the control group, the numbers of peripheral blood PMN in pre-interfere and post-interfere group were increased, there were signifi- cant differences( P < 0.01) , and there were no significant differences about the numbers of PMN between pre-interfere and post-interfere group ( P > 0.05). There was a positive correlation between the levels of ENA-78 protein and the numbers of PMN by methylprednisolone ( n = 32, r = 0.865, P<0.01). CONCLUSION: The levels of ENA-78 protein and the numbers of PMN in blood are increased in moderate and severe acute asthma children. They may be involved in the inflammation process of asthma pathogen-esis. Methylprednisolone can relieve the airway inflammation by inhibiting the expressions of ENA-78 protein.
