诱导型一氧化氮合酶在先天性肾积水中的表达及意义

摘    要:目的研究诱导型一氧化氮合酶(iNOS)在先天性肾积水肾组织的表达和分布情况,探讨它在先天性肾积水发病机制中的作用。方法采用免疫组织化学技术SABC法对36例先天性肾积水患者(病例组)、20例肾肿瘤患者正常的肾组织(对照组)进行iNOS染色,运用彩色图像分析系统测定其表达的积分光密度并作定量分析比较,同时与肾纤维化程度进行相关性分析。结果 iNOS主要表达于肾小管上皮细胞胞质、肾小球系膜细胞。积水肾组与对照组相比,iNOS表达的积分光密度明显下降,差异有统计学意义,P<0.01。积水肾组iNOS的表达与肾纤维化程度呈负相关,r=-0.649,P<0.001。结论 iNOS在积水肾表达减少加重了肾间质纤维化,iNOS所产生的一氧化氮在其发病过程中起保护作用。

Abstract:【Objcetive】 To study the role of inducible nitric oxide synthase(iNOS) in the pathogenetic mechanism of congenital hydronephrosis.【Methods】 Immunohistochemical staining(SABC method) was performed to detect the iNOS expression in 36 kidney tissues of congenital hydronephrosis and 20 normal control kidney tissues obtained from renal neoplasm.The quantitative analysis was made by image analysis technique.Correlations between the iNOS expression and the degree of renal fibrosis were investigated.【Results】 Immunoreactivity to iNOS was predominantly seen in epithelial cells of nephric tubule and mesangial cells.The iNOS expression was significantly decreased in congenital hydronephrosis group compared to normal group,P <0.01.A negative correlation was found between the iNOS expression and the degree of renal fibrosis(r =-0.649,P <0.001).【Conclusions】 The decreased expression of iNOS aggravates the degree of renal fibrosis.NO produced by iNOS may play a protective role in the pathogenesis of congenital hydronephrosis.
