PG肿瘤分泌物对PBMC活性及细胞因子分泌的影响

目的应用PG肿瘤细胞分泌的上清液培养正常外周血单个核细胞(PBMC),探讨PG肿瘤细胞分泌物对正常PBMC的活性及分泌细胞因子的影响,以进一步揭示肿瘤的免疫逃逸机制。方法将不同浓度的肿瘤细胞培养上清液与体外分离的PBMC一起培养,用MTT法检测细胞代谢活性,流式细胞术检测细胞内细胞因子分泌的变化情况。结果 PG培养上清能够提高PBMC活性,并使Th1、Th2类细胞因子表达增加。结论肿瘤上清确实能使PBMC活化,同时激活PBMC细胞分泌细胞因子。

Effects of PG cell culture supernatant on cell activity and release of cytokines of Peripheral Blood Mononuclear Cell

【Objective】 To analyze the effects of PG cells culture supernatant on the cell activity and the release of cytokines of peripheral blood mononuclear cell(PBMC),and investigate the probable mechanisms of tumor escape.【Methods】 Different concentrations of supernatants from PG cells were added into the culture system of PBMC,then the cell activity was detected by MTT and the release of cytokines was detected by Flow cytometry.【Results】 The supernatant of PG cell can improve the cell activity of PBMC.At the same time,we found that the expression of Th1,Th2 type cytokines were increased.【Conclusion】 Tumor secretions were able to make PBMC activation and activate it secreting cytokines.
