吸烟诱导的慢性支气管炎大鼠模型巨噬细胞炎性蛋白-2及髓过氧化物酶研究

摘    要:目的制备吸烟诱导的慢性支气管炎大鼠模型,研究大鼠血清髓过氧化物酶(MPO)的变化与巨噬细胞炎性蛋白-2(MIP-2)之间的关系,探讨慢性支气管炎气道炎症形成的可能机制。方法雄性Wistar大鼠随机分为二组:正常对照组、吸烟模型组。采用单纯吸烟的方法建立慢性支气管炎大鼠模型。光镜下观察支气管肺组织病理形态学改变。分析支气管肺泡灌洗液(BALF)细胞计数和分类;用ELISA法测定肺组织匀浆MIP-2质量浓度;用生化比色法测定大鼠血清MPO含量。结果模型组病理形态学改变符合人类慢性支气管炎的特点。与正常组比较,模型组血清MPO及肺组织匀浆MIP-2含量明显升高(P<0.01),模型组血清MPO与BALF中性粒细胞数,肺组织匀浆MIP-2质量浓度成显著正相关(分别为r=0.876及r=0.739;均P<0.01)。结论吸烟诱导的慢性支气管炎大鼠可能通过MIP-2介导中性粒细胞聚集,活化参与气道炎症反应。

Abstract:【Objective】To investigate the change of Myeloperoxidase(MPO) in serum and Macrophage inflammatory protein-2(MIP-2) expression in rats with chronic bronchitis induced by smoking,as well as the correlation between them and to study possible mechanisms of airway inflammation.【Methods】Male Wistar rats were randomly divided into two experiment groups:the control group,smoking model group.The rat’s model of chronic bronchitis was established by smoking purely.The histopathologic changes stained with hemotoxylin and eosin(HE) of bronchopulmonary tissues were observed under optical microscope;total cell counts and differential analysis were performed in BALF;the concentration of MIP-2 in homogenate of pulmonary tissues were measured with ELISA;Biochemical chromatometry was also used to determine the levels of MPO in serum.【Results】Pathological change of smoking model group consistent with that of chronic bronchitis.The concentration of MPO in serum and expression of MIP-2 in homogenate of pulmonary tissues in the model group significantly increased as compared with the control group(P<0.01);The concentration of MPO in serum were positively correlated with the number of neutrophil in BALF and expression of MIP-2 in homogenate of pulmonary tissues in the model group(respectively r =0.876 or r =0.739;both P<0.01).【Conclusion】MIP-2 may be involved in the process of airway inflammation through recruitment-activation of the neutrophil in rats with chronic bronchitis induced by smoking.
