黄芩苷对不同刺激剂诱导的小鼠T淋巴细胞体外活化的影响

摘    要:目的分析黄芩苷对小鼠T淋巴细胞体外活化的影响,探讨其对免疫系统的作用,阐明其免疫抑制作用机制。方法无菌分离小鼠淋巴结,制备淋巴细胞悬液,以不同终质量浓度的黄芩苷预孵育淋巴细胞4 h,再以刀豆蛋白A(Con A)或佛波醇酯类多克隆刺激剂(PDB)诱导小鼠淋巴细胞活化,利用流式细胞术分别结合双色免疫荧光单克隆抗体染色技术检测CD3 T细胞早期活化标志CD69和中期活化标志CD25分子的表达。结果黄芩苷(10、20、30 mg/L)能明显抑制Con A或PDB诱导T细胞活化分子CD69和CD25的表达,且呈浓度依赖性。结论黄芩苷能够明显抑制多克隆刺激剂Con A或PDB诱导的T细胞早期和中期活化,且呈明显的量效关系。提示黄芩苷在T细胞活化过程的信号转导中有抑制作用,从而可以通过抑制T淋巴细胞的活化,发挥免疫抑制作用,是一种潜在的免疫抑制剂。

Abstract:Objective To investigate the effect of baicalin on the activation of the mouse T lymphocytes in vitro,in order to elucidate the mechanism of the immunosuppressive effect of baicalin.Methods To isolate sterilely lymphocytes suspension from mouse lymph nodes,prepare lymphocytes which were pretreated with different concentration of baicalin for 4 h and stimulated with polyclonal activators Con A or PDB,fluorescence conjugated monoclonal antibodies and flow cytometry were used to detect the expression of CD69 and CD25 of mouse CD3 T lymphocytes activated.Results Baicalin(10,20,and 30 mg/L) could inhibit the expression of CD69 and CD25 of T lymphocytes activated by Con A or PDB in a dose-dependent manner.Conclusion Baicalin could inhibit the initial-period and mid-period activation of mouse T lymphocytes stimulated by Con A or PDB,which suggests that baicalin have the inhibitory effect on signaling pathway of T lymphocytes activation and be a potential effective immunoinhibitory agent.
