| 摘 要： | 目的 观察 8 Br cAMP对HL 6 0细胞生长、分化及其相关基因的调节。方法 组织化学、原位杂交和完整细胞RNA斑点印迹。结果 8 Br cAMP作用 2 4h ,HL 6 0细胞c fos癌基因和p2 1waf1抑癌基因表达明显升高 ,c myc癌基因表达明显降低 ;8 Br cAMP作用 48h ,HL 6 0细胞生长、增殖明显受抑制 ,并向中性粒细胞方向分化。结论 8 Br cAMP可调整HL 6 0细胞生长、分化及相关癌基因和抑癌基因的表达 ,抑制HL 6 0细胞的生长 ,促进HL 6 0细胞的分化

| Abstract: | Objective To observe the regulation of 8 Br cAMP to proliferation, differentiation and related gene expression in HL 60 cells. Methods Histochemistry,in situ hybridization and intact cell RNA dot blot techniques were used. Results After HL 60 cells were treated with 8 Br cAMP 24h, the gene expression of c fos and p 21waf1 were up regulated, the gene expression of c myc was down regulated remarkably in HL 60 cells;after 48h,the proliferation of HL 60 cells was inhibited and HL 60 cells was induced to differentiate into the granulocyte mainly. Conclusions 8 Br cAMP could regulate the gene expression related to proliferation and differentiation in HL 60 cells. |
