摘    要:目的探讨Rho激酶抑制剂(盐酸法舒地尔)对大鼠脑缺血/再灌注(I/R)后海马CA1区神经元存活的影响。方法采用Pulsinelli-Brierley四血管阻塞全脑缺血模型,缺血前30 min腹腔注射盐酸法舒地尔15mg/kg,缺血15 min后再灌注5天,断头取脑,石蜡切片,焦油紫染色,图像分析测定单位面积内焦油紫染色细胞总和,并与I/R组、生理盐水组及假手术组比较,进行形态学分析。结果盐酸法舒地尔明显减少了脑I/R所致的海马CA1区神经元损伤,与I/R组比较存活细胞数明显增多(P〈0.05)。结论盐酸法舒地尔对脑缺血/再灌注所致海马CA1区神经元损伤具有明显的保护作用。

Abstract:Objective To investigate the effect of Rho-kinase inhibitor fasudil hydrochloride on the neuronal survival of the hippocampal CA1 area following cerebral ischemia/reperfusion(I/R) in rats.Methods Transient(15 min) brain ischemia was induced by the Pulsinelli-Brierley four-vessel occlusion(4-VO) model established in Sprague Dawley rats.The rats underwent intraperitoneal injection of fasudil hydrochloride(15 mg/kg) 30 min before ischemia and subsequent 5 days of reperfusion,followed by decapitation for the removal of the brain,paraffin section and cresyl violet staining for the determination of the summation of cresyl violet stained cells and morphological analyses by light microscopy in comparison with the I/R group,normal saline group and the sham group.Result Fasudil hydrochloride significantly attenuated the I/R-induced neuronal injuries in the hippocampal CA1 area and increased the survival of the neurons as compared to the I/R group(P0.05).Conclusions Fasudil hydrochloride has a significant protective effect on the neuronal injuries induced by cerebral ischemia/reperfusion in the hippocampal CA1 area.
