摘    要:目的:探讨PI3K/Akt信号通路抑制剂LY294002对舌鳞癌细胞系Tca8113增殖和凋亡的影响及其相关机制。方法:LY294002处理Tca8113细胞,MTT法观察对细胞增殖的影响,流式细胞术检测细胞凋亡,Western blot及ELISA分析LY294002作用前后p-Akt及Akt蛋白的表达。结果:随LY294002浓度(0、10、25、50、75μmol/L)的增加及作用时间(24、48 h)的延长,实验组Tca8113细胞的增殖抑制率逐渐增大,LY294002抑制作用与浓度及作用时间呈正相关LY294002干预细胞24 h后,细胞凋亡率显著高于未干预组,p-Akt蛋白表达明显降低,而总Akt表达无显著改变。结论:LY294002可诱导Tca8113细胞凋亡,其作用机制与抑制PI3K/Akt信号通路的效应分子p-Akt的活化有关。

Abstract:Objective: To evaluate PI3K/Akt pathway inhibitor effeeting on apoptosis in human tongue squamous cancer cell line Tca8113. Methods: Tca8113 cells were treated with PI3K/Akt inhibitor LY294002. The inhibition concentrations of LY294002 for Tca8113 cells are determined by MTT assay. Cell apoptosis was analyzed by flow cytometry (FCM). The expression of phosphated Akt (p-Akt) and Akt were detected by Western blot and ELISA techniques. Results: LY294002 could inhibit the proliferation of Tca8113 ceils with a time- and dose-dependent pattern. When treated for 24 h, the apoptosis rate of Tca8113 cells was significantly higher in LY294002 groups than in control group. Moreover, the expression of p-Akt in Tca8113 cells was decreased. Conclusion: LY294002 could induce apoptosis of Tca8113 by inhibiting activity of p-Akt.
