穴位敷贴对哮喘大鼠肺脏树突状细胞表达的影响

摘    要:目的观察穴位敷贴对哮喘大鼠肺组织中树突状细胞表达水平的影响。方法将50只雄性SD大鼠随机分为正常组、模型组、地塞米松组、中药敷贴组、中药敷贴+地塞米松组,共5组,每组10只。以卵蛋白致敏并诱发哮喘模型。于大鼠颈部相当于大椎穴(G14)处,中药敷贴组予以咳喘散穴位敷贴,地塞米松组予地塞米松(1mg/kg)灌胃,中药敷贴+地塞米松组同时予咳喘散穴位敷贴和地塞米松灌胃。给药2周后,取大鼠肺组织,用免疫组化方法观察树突状细胞的表达水平。结果与正常组相比,模型组树突状细胞表达水平升高(P〈0.01);与模型组相比,穴位敷贴组、地塞米松组、穴位敷贴+地塞米松组树突状细胞表达水平均下降(P〈0.01);穴位敷贴组与地塞米松组相比差异无统计学意义(P〉0.05);与穴位敷贴+地塞米松组比较,穴位敷贴组、地塞米松组树突细胞表达更高(P〈0.05,P〈0.01)。结论穴位敷贴疗法可下调哮喘大鼠树突状细胞的表达,从而抑制肺内炎症。

Abstract:Objective To observe the effects of acupoint application on the pulmonary dendritic cells in asthma rats.Methods A total of 50 SD rats were randomized into control,model,dexamethasone(DX),acupoint application,acupoint application and dexamethasone groups,10 rats in each group.Asthma model was established by ovalbumin(OVA) sensibilization and long-term inhalation provocation.Dazhui(GV14) was selected for application of Kechuan Powder for rats acupoint application group.DX group was treated by DX(1 mg/kg) once a day,and acupoint application and DX group was given DX and acupoint application at the same time.The lung tissue was obtained after 24 hours at the end of the therapy and pathologic analysis was done through hematoxylin,eosin staining(HE staining),and the expression of DC was displayed by using immunohistochemical method.Results In comparison with control group,the expression of DC was increased significantly in model group(P〈0.01);in comparison with model group,the expression of DC was decreased in DX,acupiont application,acupoint application and DX groups(P〈0.01).No significant differences were found between DX and acupiont application groups in the expression of DC(P〉0.05).Conclusion Acupoint application therapy can down-regulate the expression of dendritic cells to suppress inflammation in lung tissues.
