左乙拉西坦对癫痫大鼠认知功能的影响研究

摘    要:目的:通过氯化锂一匹罗卡品(Li—pilo)诱导的癫痫大鼠模型,观察左乙拉西坦(LEV)对癫痫大鼠空间学习记忆能力及海马组织中突触素(SYN)表达的影响。方法:将96只SD大鼠随机分为NS组(生理盐水组),Li—pilo+LEV组(治疗组),Li-pilo组(模型组),LEV组(正常给药组),每组24R,于建模成功后7d,14d,28d通过RT—PCR方法观察各组大鼠海马组织中SYNmRNA的表达水平,并于第4周应用Morris水迷宫评价大鼠的学习记忆能力。结果:Morris水迷宫测试中Li—pilo+LEV组逃避潜伏期较Li—pilo组短(P〈0.05);Li—pilo+LEV组穿越平台次数较Li—pilo组多(P〈0.05)。PT-PCR测试各时间点NS组和Li-pilo+LEV组大鼠海马中SYNmRNA的表达量高于Li—pilo组(P〈0.05)。结论:Li—pilo点燃的癫痫大鼠学习记忆能力减退,左乙拉西坦可改善癫痫大鼠的学习记忆能力,该作用可能与其调节海马组织中SYNmRNA的表达有关。

Effects of l evetiracetam on cognition in rats with epilepsy

Abstract:Objective. To observe the effects of levetiracetam (LEV) on spatial learning and memory ability at epilepticrat and the expression of synaptophysin (SYN) in the hippocampus of rats with epilepsy induced by lithium pilocarpine (Li--pilo). Methods: 96 SD rats were randomly divided into NS group (normal saline group), Li--pito + LEV group (treatment group), Li--pilo group (model group), and LEV group (normal dose group), 24 for each group. 7th day after successful model building, expression levels of the SYNmRNA in hippocampus' tissue were detected in each group by the way of RT--PCR, and the same way was used for the detection of expression at the 14th and 28th day. In the 4th week, to evaluate the ability of learning and memo- ry in each group rats, by the Morris water maze. Results. The Morris water maze test results show that escape latency of Li- pilo + LEV group was shorter than that of Li--pilo group in the 1Morris water maze test (P 〈 0.05) ; the times of crossing the location of platform of Li--pilo + LEV group was more than Li--pilo group (P 〈 0. 05) ; at each time point, the expression of SYNmRNA in the rat hippocampus of NS and Li--pi- loq-LEV group was higher than Li--pilo group (P 〈 0.05). Conclusion: Learning and memory abilities de- clined in epilepsy rats kindled by Li--pilo; levetiracetam can improve learning and memory ability of rats with epilepsy, and the effect may be related tO its regulation of SYNmRNA expression in hippoeampal tissue.
