经门静脉输注供者特异性血管内皮细胞诱导豚鼠一大鼠异种移植免疫耐受的实验研究

摘    要:为探讨异种移植免疫耐受的可能性,以豚鼠一大鼠异位心脏移植超急排模型,在CVF(30U/kg)及环磷酸胺(CyP)覆盖下将供者特异性血管内皮细胞以4.0X106/只经受者门静脉输注,2周后行颈部心脏移植使移植供心存活时间获得延长(44.8~64.2min),同时血管内皮细胞溶解率下降。环磷酰胺及脾切除有一定延长移植物存活的作用(25.9~43.3min,30~47.2min),而非特异性血管内皮细胞无此作用。提示经门静脉输注供者特异性豚鼠血管内皮细胞可诱导大鼠产生一定程度的免疫耐受,这种耐受的产生机制尚需进一步研究。

Abstract:Induction of allograft tolerance has been successful in many animal ~is and concordant xenograft also, but disconlant xenon has never been attempted with success. In this stUdy,donor-- speCific gUinea -- pig arterial endothelial cells were isolated and amplified by in vitID cul~,then injected (4.0 x lab/rat) into portal vein after recipient ratS were pretreated with Cobra Venom Factor (CVF, 30U/kg) along or combined with Cyclophosphamide (CyP, 20mg/kg). Guinea -- pig hear'tSwere hansplanted heterologically. Caldiac survival time of treated rats with specific ECs was much longerthan that of control ratS (46. I t 8.smin vs. 16.4 t 4.7min, P < 0.05), while using CyP addihonallywas more effective (survial bine 54.5 t 9. 7 min, P < 0.05). SPlenectomy and CyP could prolong thesurvival time in hide (36.4 i 8 .7lain and 38 .6 t 8 .6min). Non -- specific ECs couldn't prolong graftsurvival time. A close negative correlation of ECs lytic ratio with graft survival time in each group wasobserved. Histological stud), found no appal.ent difference among all groups qUalitatively and qUantitahvely at the end of rejection. The results suggest that injecting of donor -- specific ECs into recipientrats'portal vein could lead to xenogiaft tolerance between gUniea -- pick and rats but it couldn't preventHAR completely. ECs lyhc ratio was helpful in study of AR in vitro.
