曲古菌素A对视网膜母细胞瘤Hxo-rb44细胞凋亡及Survivin、c-Myc表达的影响

摘    要:目的:探讨曲古菌素A(TSA)对人视网膜母细胞瘤(Rb)细胞株Hxo-rb44细胞凋亡和Survivin、c-Myc表达的影响.方法:将人Rb细胞株Hxo-rb44进行培养传代,流式细胞仪检测不同浓度的TSA作用不同时间对Hxorb44细胞周期和凋亡的影响,用Western blot法检测凋亡过程中Survivin和c-Myc蛋白表达的变化.结果;TSA可明显诱导细胞发生G2期阻滞及细胞凋亡,而且与药物作用时间呈依赖关系(r=0,97,P=0,02);Western blot法可以观察到Survivin和c-Myc的表达水平随着TSA作用时间的延长而下降(P均<0,01).结论:TSA可以有效诱导人Rb细胞株Hxo-rb44细胞周期阻滞和凋亡,其作用机制与抑制Survivin和c-Myc表达有关.

Abstract:Aim: To investigate the effect of trichostatin A (TSA) on apoptosis and expression of Survivin and c-Myc of retinoblastoma cell Hxo-rb44.Methods:The retinoblastoma cell Hxo-rb44 was chosen in this experiment. The cell cycle arrest and apoptosis of Hxo-rb44 cell were assayed with flow cytometry. Expression of Survivin and c-Myc was detected by Western blot. Results: TSA obviously induced cell cycle arrest and apoptosis in time-dependent manner (r=0.97,P= 0.02 ). The expression of Survivin and c-Myc decreased with TSA incubation time (P<0.01).Conclusion: TSA can effectively induce cell cycle arrest and apoptosis in Hxo-rb44 cell, which may associate with TSA suppressing the expression of survivin and c-Myc.
