川芎嗪对离体灌注大鼠肝脏缺氧的保护作用

目的:探讨川芎嗪对离体灌注大鼠肝脏组织在缺氧情况下的保护作用。方法:将离体大鼠肝脏的灌注模型分为缺氧对照组和缺氧川芎嗪保护组（160mg川芎嗪）。分别测量基础对照、缺氧和缺氧恢复3个阶段的血红蛋白（Hb）浓度和氧合血红蛋白（HbO2）浓度,并且利用计算机辅助扫描系统得出Hb和HbO2浓度的空间分布图。结果:缺氧可使Hb浓度增加,川芎嗪未改变由缺氧引起Hb增加。缺氧可以造成HbO2浓度降低,川芎嗪可以显著减小HbO2浓度减低的幅度（P<0.01）。所测的三维空间分布图显示:缺氧可导致肝脏组织中的功能区破坏,川芎嗪对功能区起到保护作用。结论:中药川芎嗪对离体灌注的大鼠肝脏有明显的抗缺氧保护作用。

The protective effect of Ligustrazine against hypoxia on isolated perfused rat liver

Objective:To investigate the influence of hypoxia on isolated perfused rat liver and the protective effect of Ligustrazine. Methods:Hypoxia state in the isolated perfused rat liver model was established. In group A,Ligustrazine was not added into the perfusion system; in group B, 160 mg Ligustrazine was added into the perfusion system. Using EMPHO SSK and its scanning system, concentrations of hemoglobin and the oxygenation of the hemoglobin in the isolated perfused rat liver at the baseline stage, hypoxia stage and reoxygenation stage were measured, and the 3-dimension images of them also obtained. Results:Hypoxia could elevate the concentration of hemoglobin in the liver, but reduce the oxygenation of the hemoglobin, and it could damage the functional units (illustrated by the 3-D images) of the liver. Ligustrazine did not interfere the intra-capillary hemoglobin concentration increase, but improve the intra-capillary hemoglobin oxygen saturation when the hypoxia happened. From the 3-D images,Ligustrazine was seen to protect the functional units of liver tissue from the damage of hypoxia. Conclusion: Ligustrazine could protect the siolated perfused rat liver from the damge of the hypoxia.
