摘    要:目的:通过研究骨髓增生异常综合征患者血清造血抑制活性,探讨致MDS无效造血的可溶性因素。方法:以甲基纤维素半固体细胞培养法,观察患者血清对正常骨髓BFU-E,CFU-GM的抑制作用,及对单个核细胞素集落生成的刺激作用;以DNA末端标记技术,检测1例MDS患者血清与正常骨髓单个核细胞共育后的凋亡细胞。

Abstract:Objective: To explore the soluable factors which result in ineffective hematopoiesis through the investigation of inhibitory activities of MDS sera on hematopoietic progenitor cells. Methods: Suppresion of granulocyte-macrophage progenitors(CFU-GM) and erythroid burst-forming units(BFU-E) and stimulation of normal BMMNCs colony-formating by sera from patiens with MDS were examined in a semisolid methycellulose culture system in vitro.Normal BMMNCs incubated with 1 patient's serum were assayed for Progammed Cell Death(PCD) by the in situend labeling (ISEL) technique. Results: 6 of 15 sera from patients suppressed the colony formation of BFU-E,2 of 15 suppressed the colony formation of CFU-GM,while 11 of 15 sera stimulated the autogenous colony-formation,In fluid culture system 1 patient's serum promoted the PCD of normal BMMNCs. Conclusion: Some soluble factors might be responsible for the abnormal regulation of hematopoiesis,even for the unique apoptosis mechanism in MDS.
