摘    要:目的:观察HD02对慢性前脑缺血大鼠神经干细胞增殖分化的变化。方法:结扎双侧颈总动脉制备慢性前脑缺血大鼠模型,采用免疫单标和双标方法检测成体大鼠神经干细胞及分化的功能细胞。给予模型大鼠HD02水煎剂灌胃,分别于造模后15,30d观察神经干细胞增殖分化的变化。结果:与正常对照组相比,慢性前脑缺血大鼠大脑皮质、室周区、海马BrdU阳性细胞数和BrdU+NF200,BrdU+NeuroD,BrdU+GFAP免疫荧光双标细胞数于造模后15d明显增多(P<0.01),但造模后30dBrdU阳性细胞数和BrdU+NF200,BrdU+NeuroD,BrdU+GFAP免疫荧光双标细胞数与正常对照组相比差异无显著性意义(P>0.05);造模后30d,HD02组BrdU阳性细胞数和BrdU+NF200,BrdU+NeuroD,BrdU+GFAP免疫荧光双标细胞数较慢性前脑缺血组明显增多(P<0.01)。结论:HD02可显著增加慢性前脑缺血大鼠神经干细胞增殖分化能力

Abstract:AIM:To explore the effects of HD02on differentiation and proliferation of neural stem cells in chronic cerebra l ischemia rats.METHODS:Sprague-Dawley rats were subjected to permanent bilateral carotid arteries ligation(BCAL)for 2months.Single and double immun ohis-tochemical stainings were used to id entify neural stem cells and differe ntiated neural cells in aged rats which were t reated with HD02(5g /kg)at 15and 30d of ischemica respectively.RESULTS:Compared with the normal control gro up,the number of BrdU immunofluorescence positive cells and BrdU +NF200,BrdU+NeuroD,Br-dU +GFAP double-immunostaining of c erebral cortex,subventricular zon e,hippocampus in chronic cerebral isc hemic group was markedly increased a t 15d after ischemia(P<0.01),but at 30d after ischemia there were no sig-nificant differences between the normal control group and chronic cereb ral is-chemic group(P>0.05).After the treatment of HD02,the num ber of BrdU and double-immunostaining of BrdU +NF200,BrdU +NeuroD and BrdU +GFAP was significantly increased as compared with the chronic cerebral i s-chemia group at 30d after ischemia re spectively(P<0.01).
