受体T淋巴细胞克隆的建立及其抗原特异性的检测

目的　建立受体T淋巴细胞克隆后 ,进一步分析检测该克隆针对供体的抗原特异性。方法　供体鼠脾细胞免疫受体鼠 ,再分离、纯化受体鼠T细胞 ,克隆受体T细胞。将动物分组免疫 ,对各组受体作电镜观察淋巴细胞超微结构、MTT法测定淋巴细胞增生试验以及流式细胞分析T细胞亚群的变化 ,分析检测该克隆针对供体的抗原特异性。结果　①受体针对供体抗原特异性T淋巴细胞克隆稳定建立 ;②经免疫组的受体T细胞超微结构改变显著 ,淋巴细胞增生试验及流式细胞分析结果与对照组的差异有显著性意义 (t>t0 0 5)。结论　成熟T淋巴细胞 ,在一定条件下仍可发生分裂和增生 ,形成克隆。通过供体脾细胞抗原特异性刺激 ,检测证明最终筛选出的T细胞克隆具有针对供体的抗原特异性

Establishment of T lymphocyte clone of recipient and analysis on its donor-antigen-specificity

Objective The aim of this study was to analyze the antigen-specificity of the clone after the recipient-against-donor antigen-specific T lymphocytes clones were formed.Methods The recipient rats were immunized by the donor rats′spleencytes,then their T cells were isolated and purified.The T cells were cultured by adding raise cells,ConA or IL-2 at different concentrations and formed the T cell clone in the last.The changes of the ultrastructure,cell multiplication and subgroup of leukemic cells were investigated when the rats were immuned.Results A stable recipient-against-donor antigen-specific T cell clone was established;statistical data also verified that the difference between the immune groups and the comparison group was very significant (t>t 0.05).Conclusion Mature T cell is able to proliferate to form clone in a suitable condition.Through stimulating the donor spleen cell antigen specificity the experiment proved that the finally-screened T-cell clone has an antigen-specificity to the donor.
