急性胃肠炎暴发流行的病原学研究

目的查明承德市急性胃肠炎暴发原因,确定病原体。方法采集患者粪便标本,采用ELISA方法检测腹泻病人粪便标本病毒抗原,对ELISA结果阳性标本提取病毒核酸并通过特异性引物进行PCR扩增、测序并与参考株做同源性分析,结合临床症状及流行病学特征,确定发病原因。结果 21份粪便标本中,6份标本诺如病毒抗原阳性,核酸扩增和测序表明为诺如病毒GII型,序列分析表明,扩增片段序列与GII/12亚型诺如病毒序列相似性最高。结论证实该起胃肠炎由诺如病毒GII型引起。

The etiological study on the outbreak of acute gastroenteritis in Chengde city of Hebei province

Objective To identify the pathogen resulting in an outbreak of acute gastroenteritis in Chengde city of Hebei province.Methods The stool specimens of patients with acute gastroenteritis were collected and the possible viral pathogen was detected by ELISA.Then the viral nucleic acid was extracted from the positive specimens and amplified with specific primers by PCR,which was sequenced and analyzed for its homology according to reference viral strain,finaly,the cause of disease was determine according to the results of laboratory detection combined with patients’ symptoms and epidemiological features.Results Among 21 stool specimens,6 specimens were positive for norovirus antigen,which belonged to type GII of norovirus identified by nucleic acid amplification and sequencing.The results of gene sequence analysis showed that the sequence of amplification fragment was highly similar to that of GII/12 subtype of norovirus.Conclusion The laboratory results confirm that the outbreak of acute gastroenteritis in Chengde city of Hebei province is caused by GII subtype of norovirus.
