间歇低氧幼鼠8-异前列腺素F2α变化及芹菜素的干预效果

摘    要:目的:观察间歇低氧幼鼠血清及海马8-异前列腺素F2α(8-ISO—PGF2α)的变化及芹菜素的干预效果。方法:取3~4周龄健康雄性SPF级SD幼鼠32只,随机分为间歇低氧组(IH)、空气对照组(C)、芹菜素十二甲基亚矾组(Q)和二甲基亚砜组(R),每组8只,以间歇低氧舱建立间歇低氧幼鼠模型,99.99%的纯氛和99.50%的医用氧交替输入舱内,90s为一次低氧-复氧循环;间歇低氧2周后,ELISA法测定各组血清及海马8-ISO—PGF2α水平。结果:血清中,IH组和R组8-ISO—PGF2α的含量分别高于C组和Q组(均P〈0.01),而C组与Q组比较差异无显著性(均P〉0.05),IH组与R组比较差异无显著性(均P〉0.05);在海马中,结果类似;血清和海马组织中8-ISO—PGF2α含量呈正相关(r=0.818,P〈0.01)。结论:间歇低氧后幼鼠血清及海马中8-ISO—PGF2α表达均升高,芹菜素可降低血清和海马中的8-ISO—PGF2α水平,在治疗间歇低氧造成的损伤中具有应用前景。

Abstract:Objective: To detect the changes of 8-iso-prostaglandin F2α (8-ISO-EGF2α) in serum and the hippocampus and to observe the intervention effect of apigenin in immature rats exposing to intermittent hypoxic. Methods: Thirty two 3-4 week-aged SDrats were randomly divided into intermittent hypoxia (IH) group,air control(C) group,apigenin+ DMSO (Q) group and DMSO control(R) group. Each group had eight rats. Intermittent hypoxia model was induced by an intermittent hypoxia cabin and the cabin was flushed with 99.99% nitrogen alternating with 99.5% oxygen. The cycle of hypoxia- reoxygenation was 90 s. After 2 weeks, the serum and hippocampus levels of 8-ISO-Prostaglandin F were tested by ELISA. Results: In the serum, the expression of 8-ISO-FGF in IH and R groups were significant higher than that in C and Q groups (F〈0.01, respectively). There were no difference between C and Q groups (P〉0.05), and no difference between IH and R groups (P〉0.05). The similar results were found in hippocampus. Serum 8-ISO-PGF2α and hippocampus 8-ISO-PGF were positively correlated (r =0.818, P〈0.01). Conclusion: The levels of 8-iso-PGF2α, increases in both serum and hippocampus of immature rats with IH and the levels of 8-iso-FGF in serum can reflect indirectly the degree of oxidative stress in hippocampus. Apigenin can reduce the level of 8-iso-PGF in serum and hippocampus. Therefore, it has plication prospect in treatment of injuries which induced by IH.
