地黄饮子孵育BMSCs移植对脑梗死大鼠MMP-9和TIMP-1的影响

摘    要:目的研究地黄饮子孵育骨髓间充质干细胞(bone marrow stromal stem cells,BMSCs)移植对脑梗死大鼠脑组织中基质金属蛋白酶-9(MMP-9)和金属蛋白酶组织抑制剂-1(TIMP-1)的影响,探讨其对大脑的保护作用。方法将健康SPF级Wistar大鼠随机分为假手术组、模型组、BMSCs组、维甲酸组和地黄饮子组,用线栓法制成Wistar大鼠大脑中动脉缺血模型(MCAO),术后24 h假手术组、模型组大鼠经尾静脉注射生理盐水,其余各组大鼠经尾静脉输入相应药物孵育的BMSCs。移植后7 d,14 d断头取脑,用免疫组化法检测脑组织中MMP-9和TIMP-1的表达情况。结果与假手术组比较,模型组MMP-9表达明显增加(P〈0.05);地黄饮子组、维甲酸组、BMSCs组与模型组比较,MMP-9的表达均降低(P〈0.05),但仍高于假手术组。模型组TIMP-1的表达明显降低(P〈0.05),各治疗组均增高(P〈0.05)。结论地黄饮子孵育BMSCs可通过调节MMP-9与TIMP-1的表达,进而发挥脑保护作用。

Abstract:Objective To observe the effect of transplanted bone marrow stromal stem cells incubated with Di- huangyinzi on the expression of MMP-9 and TIMP-1 in the tissue of cerebral infarction rats and discuss its cerebral protective role. Methods The healthy SPF Wistar rats were randomized into pseudo-operation group, model group, BMSCs group, RA group and Dihuangyinzi group. Permanent models of middle cerebral artery occlusion by tight line were made. 24 h after operation, the pseudo-operation group and model group were injected with isotonic Na chloride through vena caudalis, while the other groups were injected BMSCs incubated with Dihuangyinzi, etc. On the seventh day and fourteenth day after operation, the rats were decapitated to get the brains which were exa-mined with immunohistochemical method to detect the expressions of MMP-9 and TIMP-1. Results Compared with the psuedo-operation group, MMP-9 increased markedly in model group( P〈0.05 ) ; the expression of other groups lies between the pseudo-operation group and model group(P〈0.05). There was a significant TIMP-1 in- crease in treatment groups compared with model group(P〈0.05 ). Conclusion BMSCs incubated with Dihuang- yinzi can protect the brain through adjusting the expressions of MMP-9 and TIMP-1.
