南瓜蛋白联合吉非替尼对人胰腺癌BxPC-3细胞的抑制作用

摘    要:目的研究南瓜蛋白(cucurmosin,CUS)联合表皮生长因子受体(epidermal growth factor receptor,EGFR)酪氨酸激酶抑制剂吉非替尼(Gefitinib,GEF)对人胰腺癌细胞株Bx-PC-3的生长抑制作用。方法采用SRB法测定CUS与GEF联用对体外培养的BxPC-3细胞的增殖抑制作用;应用集落形成实验测定CUS与GEF联用对体外培养的BxPC-3细胞集落形成的影响;以Western blot技术检测CUS与GEF联用对体外培养的BxPC-3细胞EGFR的影响。结果 CUS联用GEF对BxPC-3细胞的增殖抑制率大于单用药物,q>0.85;CUS联用GEF对BxPC-3细胞的集落形成抑制率大于单用药物,q>1.15;CUS联合GEF干预BxPC-3细胞可明显下调其EGFR蛋白表达,q>2.0。结论 CUS联合GEF对人胰腺癌BxPC-3细胞的生长抑制具有协同作用。

Abstract:Aim To investigate the effects of cucurmosin(CUS) combined with EGFR Tyrosine Kinase Inhibitor,gefitinib(GEF),on human pancreatic cancer cells.Methods SRB assay was used to detect proliferation inhibition in human pancreatic cancer cell lines BxPC-3 in vitro after the treatment of CUS combined with GEF.Colony forming assay was used to observe the colony forming efficiency in BxPC-3 cells treated by CUS combined with GEF.Western blot was used to analyze the proteins expressions of EGFR in BxPC-3 cells after the treatment of CUS combined with GEF.Results The inhibition rate of CUS combined with GEF was higher than CUS or GEF alone,q>0.85.The colony formation inhibition rate of the combined treatment group was higher than the groups of CUS or GEF alone,q>1.15.The combined treatment group down-regulated EGFR protein significantly,q>2.0.Conclusion CUS combined with GEF shows a synergistic effect on cells growth inhibition in BxPC-3 cells in vitro.
