黄角颗粒对脑缺血再灌注损伤大鼠JAK2/STAT3信号通路的影响

摘    要:目的:研究黄角颗粒对脑缺血再灌注损伤大鼠JAK2/STAT3信号通路的影响。方法:采用线栓法构建大鼠脑缺血再灌注损伤模型,取30只健康雄性SD大鼠随机分为假手术组、模型组和黄角颗粒组,并按分组给予相应处理。采用Zea Longa评分法对各组大鼠进行神经功能评分,TTC染色检测各组大鼠脑梗死体积百分比,HE染色观察各组大鼠脑组织病理形态,TUNEL染色法检测各组大鼠脑细胞凋亡率,Western blot检测各组大鼠脑组织中p-JAK2和p-STAT3的蛋白水平。结果:与假手术组相比,模型组大鼠的神经功能缺损程度和神经细胞损伤程度明显加重(P0.05);脑梗死体积百分比和脑细胞凋亡率显著上升(P0.05);脑组织中p-JAK2和p-STAT3的蛋白水平显著上调(P0.05)。与模型组相比,黄角颗粒组大鼠的神经功能缺损程度和神经细胞损伤程度明显减轻(P0.05);脑梗死体积百分比和脑细胞凋亡率显著下降(P0.05);脑组织中p-JAK2和p-STAT3的蛋白水平显著下调(P0.05)。结论:黄角颗粒对脑缺血再灌注损伤大鼠的保护作用可能与抑制JAK2/STAT3信号通路的激活相关。

Abstract:AIM:To investigate the effect of Huangjiao granule on JAK2/STAT3 signaling pathway in rats with cerebral ischemia/reperfusion injury. METHODS:The rat model of cerebral ischemia/reperfusion injury was established by suture method. The rats were divided into sham operation group, model group and Huangjiao granule group. Zea Longa scoring was used to evaluate the neurological function of the rats in each group. The percentage of cerebral infarction volume in each group was measured by TTC staining. The pathological changes of the brain tissue in each group were observed by HE staining. TUNEL staining was used to detect cell apoptosis of brain tissue in each group. The protein levels of p-JAK2 and p-STAT3 in the brain tissues in each group were determined by Western blot. RESULTS:Compared with the sham operation group, the degree of neurological deficit and the degree of neuronal damage were significantly increased in model group (P<0.05). The percentage of cerebral infarction volume and the apoptotic rate of the brain tissue were significantly increased in model group (P<0.05). The protein levels of p-JAK2 and p-STAT3 in the brain tissues of model group were significantly up-regulated (P<0.05). Compared with model group, the degree of neurological deficit and the degree of neuronal damage were significantly decreased in Huangjiao granule group (P<0.05). The percentage of cerebral infarction volume and the apoptotic rate of the brain tissues were significantly decreased in Huangjiao granule group (P<0.05). The protein levels of p-JAK2 and p-STAT3 in the brain tissues of Huangjiao granule group were significantly down-regulated (P<0.05). CONCLUSION:The protective effect of Huangjiao granule on rats with cerebral ischemia/reperfusion injury may be related to the inhibition of JAK2/STAT3 signaling pathway activation.
