2010年青海省流感病毒病原学监测及H3亚型HA1基因序列分析

目的监测青海省2010年季节性流感病毒型与亚型分布,并了解2010年部分A/H3N2分离株血凝素(HA)基因变异情况。方法采集全省各监测哨点医院鼻咽双拭子标本,接种MDCK细胞,采用标准抗血清鉴定阳性分离株型与亚型,并对2010年部分H3亚型流感病毒进行HA基因测序,分析HA基因变异情况。结果2010年全省哨点医院送检标本中共分离63株流感病毒,其中42株为A/H3N2,B型15株,AH1N1流感病毒6株,型与亚型有明显分布强弱特征。选取10株H3N2亚型分离株进行HA序列分析,发现与同期南京分离株接近,与WHO2010年度疫苗推荐株相距甚远,不在同一分支。结论 2010年青海省流行的H3N2亚型流感病毒的抗原性和基因特性发生了改变。

Pathogen surveillance of influenza virus and H3 isoforms HA1 gene sequencing at 2010 in Qinghai Province

Objective To understand the type and subtype distribution of seasonality influenza virus and hemagglutinin(HA) gene variation of subtype A/H3N2 at 2010 in Qinghai Province.Method Collected nasopharynx swabs specimens in sentinel hospitals,inoculated MDCK cells and then identified by hemagglutination inhibition(HI) method.Part of H3 subtype virus in 2010 were carried out HA gene sequencing,and analyzed the HA genovariation.Results It was shown that 63 strains influenza virus were isolated from the specimens,among them,42 strains were A/H3N2,15 strains were type B and 6 were A/H1N1.The distribution of type and subtype were significantly different.After HA gene sequence analysis of H3N2,we knew that the sequences of 10 strains which isolated form Qinghai Province in 2010 were similar to that of isolated from Nanjing and far from vaccine strains recommended by WHO at 2010,it was not in the same branch of a tree.Conclusions The antigenic and genetic characteristics of H3N2 isoforms influenza virus in Qinghai in 2010 had changed.
