摘    要:目的对Trizol法中不同组织研磨方法进行比较,优化新生鼠脑总RNA提取方法。方法取新生24h小鼠脑组织,分别采用常规玻璃组织研磨器(常规研磨法)、剪刀剪碎法、液氮研钵研磨(液氮研磨法)3种方法对组织匀浆进行分离,采用Trizol试剂提取总RNA,紫外分光法测定浓度和纯度,凝胶电泳验证完整性。结果 3种方法均可以提取出总RNA,但相对剪碎法和常规研磨法,液氮研磨法提取的总RNA浓度、纯度及完整性更佳(P<0.05)。结论液氮研磨法虽然对实验条件有一定要求,但仍然是目前最为可靠的脑组织总RNA提取方法,可以完全满足后续实验需要。

Trizol extraction of total RNA from neonatal mouse brain tissue for optimization

Abstract:Objective To explore the best one of three grinding methods in the extraction of RNA from neonatal mouse brain tissue by Trizol.Methods The tissue homogenate of neonatal mouse brain was isolated via three grinding methods,regular way,scissors way and liquid nitrogen way.Total RNA was isolated with the Trizol,the concentration and purity were determined by ultraviolet spectrophotometry,and the integrity was determined by gel electrophoresis.Results The total RNA could be isolated by three grinding methods,but the best one was liquid nitrogen way(P<0.05).Conclusion The liquid nitrogen grinding requires the stricter experiment condition,but it is the most reliable isolation of RNA.
