摘    要:目的:建立以反相高效液相色谱法测定法半夏中γ-氨基丁酸含量的方法。方法:在碱性条件下,氨基酸与异硫氰酸苯酯反应,生成异硫氰酸苯-氨基酸的衍生物;采用Venusil-AA氨基酸分析柱(250mm×4.6mm,5μm)、二元梯度洗脱方式测定样品含量,流动相A为0.05mol.L-1醋酸钠溶液(pH6.5),流动相B为乙腈-水(80∶20),检测波长为248nm。结果:γ-氨基丁酸检测浓度在2.056~12.336μg·mL-1范围内与峰面积积分值呈较好的线性关系;加样回收率为99.0%,RSD=0.92(n=6)。结论:本方法简便、准确、重现性好,可用于法半夏的质量控制。

Abstract:OBJECTIVE: To determine the content of γ- aminobutyric acid in Rhizoma Pinelliae Praeparatum.METHO, DS: In alkaline condition, amino acids reacted with phenyl isothiocyanate and resulted in the derivatives of benzene isothiocyanate - amino acid complex. A Venusil-AA amino acid analysis column(250 mm ×4.6 mm, 5μm) was used and the absorbance at 248 nm was measured. The mobile phase A is 0.05 mol· L ^-1 sodium acetate solution (pH6.5) and the mobile phase B consisted of acetonitrile-water (80 : 20). RESULTS: The linear range of γ- aminobutyric acid was from 2.056μg·mL^-1 to 12.336μg· mL^-1, and the recovery was 99.0% (RSD = 0.92, n = 6). CONCLUSION: This method is simple, accurate and reproducible, and it can be used for quality control of Rhizoma Pinelliae Praeparatum.
