乙酰白藜芦醇的制备及其含量的高效液相色谱法测定

摘    要:目的:以白藜芦醇为原料制备乙酰白藜芦醇,并建立高效液相色谱法测定其含量。方法应用白藜芦醇与醋酸酐120℃加热反应制取乙酰白藜芦醇。色谱柱采用inertsil ODS-SP柱(150 mm ×4.5 mm,5μm),流动相为乙腈-水(45∶55,V/V),流速为1.0 ml/min,检测波长为300 nm,进样量为20μl,柱温为25℃。结果反应获得高纯度的乙酰白藜芦醇,产率达90%;乙酰白藜芦醇质量浓度在20.0~400.0μg/ml范围内与峰面积线性关系良好(r=0.9999),平均回收率为99.82%,RSD为0.89%(n=6)。结论该合成方法简单易行,可用于乙酰白藜芦醇的制备;建立的检测条件准确、简便,精密度高,可用于其含量测定。

Abstract:Objective To prepare of acetyl resveratrol and establish a high performance liquid chromatogra-phy(HPLC) method for its content determination .Methods Resveratrol and acetic anhydride as raw materials ,the reaction temperature was 120 ℃.The inertsil ODS-SP column(150 mm ×4.5 mm,5 μm) was used and the mobile phase was CH3CN-H2O(45∶55,V/V),the flow rate was 1.0 ml/min.The detection wavelength was 300 nm.The col-umn temperature was 25℃and the injective volume was 20μl.Results The reaction get a high purity of acetyl res-veratrol ,the yield was 90%.A good linear relationship was observed in the concentration range of 20.0~400.0μg/ml, with the correlation coefficient of 0.999 9 .The average recovery rate was 99.82% and RSD was 0.89%( n=6 ) . Conclusion The synthetic method is proved to be simple and the determination method was accurate and sensitive with high precision ,which can be applicable for the preparation of acetyl resveratrol and its content assay .
